英文摘要 | The specific activity of γ- glutamyl transpeptidase in the plasma membrane fraction of rat liver, measured by spectrophotometry is found to be enhanced by the addition of a non-ionic detergent, Triton X-100. The optimal concentration of Trition x-100 for the measurement of the γ-glutamyl transpeptidase activity is 0.1%. The specific activity of γ-glutamyl transpeptidase in 0.1% Triton X-100 treated membrane fractions is 69.73 ��2.35 nmol/min/mg protein, and the specific activity of the enzyme in membrane fractions without Triton X-100 treatment is 42.65 �� 0.60 nmol/min/mg protein this represents a 1.6-fold enhancement of the γ-glutamyl transpeptidase activity upon surfactant solubilization (p<0.05). Both Km and Vmax of the enzyme are altered by the addition of Triton X-100. The γ -glutamyl transpeptidase activities in cellular homogenate and nuclear envelope of rat liver are also assessed by the same method. |
