中文摘要 | Studies on the chronic administration of iron (II), manganese (II) and 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) were carried out in mice to examine their mode of action in non-neural tissues. Results indicate that the treatments increased cardiac protein levels but decreased hepatic, renal, splenic and lung protein levels relative to the controls. These changes were significant (P<0.05) except in the heart and spleen after MPTP treatment. The treatments also resulted in significant (P<0.05) inhibition of hepatic triacylglycerol (TG) lipase activity compared to the control. Rehabilitation of the treated animals with diethyldithiocarbamate (DDC) could not reverse the effects of Fe(II), Mn(II) and MPTP in the heart and lungs. This pharmacological agent, however, reversed the effects of treatment in the kidney, liver and spleen. DDC-induced reversal was most pronounced after MPTP treatment where significant (P<0.05) increases in the protein levels relative to the controls were observed. However, DDC therapy had no obvious effect on hepatic TG lipase activity. Conversely, L-Dopa therapy reversed the inhibition caused by Fe(II), Mn(II) and MPTP on the TG lipase activity.Similarly, L-Dopa reversed the effect of Fe(II) < Mn(II) and MPTP in the liver and spleen. These results suggest differences in the mode of action of Fe(II), Mn(II) and MPTP in spite of many apparent similarities. |
