中文摘要 | 由胡蘿蔔(Daucus carota L. cv. Shinkuroda Gosun)及水稻(Oryza sativa L. Taipei 309)所育出之枝及根的懸浮細胞,分別施以0%到25% Polyethylene glycol (PEG) 8000水分逆境處理,以研究不同部位培植體所育出之細胞在水分逆境中之生理差異。胡蘿蔔枝細胞在5% PEG中生長較快,水稻枝及根細胞在逆境中之部分時期生長較佳。胡蘿蔔與水稻枝細胞在逆境處理初期,其蛋白質含量均較根細胞為高。Proline含量在胡蘿蔔枝細胞中均較根細胞含量為高。經由SDS-PAGE分析兩種植物之根與枝細胞在逆境強時蛋白質會改變,兩種植物根細胞澱粉含量均較枝細胞為高,且枝細胞之蔗糖、果糖及葡萄糖在各水分逆境中均較根細胞為高。部分蔗糖、果糖、葡萄糖及蛋白質在5% PEG中有突增現象,可能與生長中之滲透調節有關,結果顯示來自不同部位之細胞,在水分逆境中具有不同之生理反應。 |
英文摘要 | The physiological differences of stoot-derived and root-derived suspension cultured cells of carrot (Daucus carota L. cv. Shinkuroda Gosun) and rice (Oryza sativa L. Taipei 309) in concentrations between 0% and 25% polyethylene glycol (PEG) 8000-induced water stress were studied. The carrot shoot cellsglew faster in 5% PEG than the other stress. The rice shoot and rootcells glew faster in water stress for some periods. Both kinds of shoot cells had more protein content than root cells in the initialtreatment. By the analysis of SDS-PAGE, the shoot and root cell sprotein of the two plants will change in higher stress condition. Both kinds of root cells had more starch content than the shoot cells. And the shoot cells had higher sucrose, fructose and glucose content than the root cells in water stress. Substantial osmotic adjustment with higher growth rate in 5% PEG was observued, due mainly to theincrease of sucrose, glucose, fructose and protein. The resultsindicated that the cells come from different sources had different physiological characteristics in water stress. |
