英文摘要 | Mycoplasma infections are important because they cause economic losses in commercial animal production. A universal two-step polymerase chain reaction (PCR) was used to detect mycoplasma infections. Samples derived from ring-necked pheasants and cell lines were amplified by two-step PCR and further digested with restriction enzymes, Taq I and Eco RI. Restriction fragment profiles generated by Taq I showed herterogeneity among species. The two-step PCR allowed the sensitive detection of mycoplasmas when te quantity of mycoplasma in the tissue or cells is low. The results revealed that reamplification with nested primers increased its sensitivity. Therefore, the two-step PCR provided a rapid, sensitive, and specific approach to detect mycoplasma contamination in cell cultures and to aid in the diagnosis of mycoplasma infections. |
