中文摘要 | 本研究乃為探討異體來源之人類核糖核酸萃取物經小白鼠腹腔注射後,移植性免疫能力之效應,ICR種系小白鼠被選配分為五大組,第一組為正常控制組,第二組經五葛雷全身輻射組,第三組為經核糖核酸前處理及輻射合併組,第四組為核糖核酸及輻射合併組,第五組為實驗控制組乃單獨核糖核酸十次處理,iRNA每週給藥兩次,每次劑量為每公斤體重0.16毫克,在經處理後分別在第1, 7, 14, 21天,每組犧牲動物五隻分析之,脾臟重量在輻射後受到明顯抑制,而經iRNA處理後能夠幫助其恢復脾臟重量,而在胸腺重量經輻射後有明顯的失重,然而在iRNA單獨處理下有輕微的增加,而對於骨髓細胞在輻射後各處理組皆有恢復的作用,這些結果顯示人類脾臟核糖核酸萃取物似乎能夠幫助經輻射後動物免疫器官之免疫能力及巨噬細胞吞噬能力的恢復。 |
英文摘要 | To investigate the effectiveness on transplantation immunity of Human spleen RNA from xenogenic sources by intraperitoneum injection in irradiated mice. ICR strain male mice were chosen and divided into five groups. group A was normal control, and group B was treated with whole body exposure of 5Gy γ-irradiation (RT), and Group C was treated with RT after being treated with iRNA. Group D was treated with RT and iRNA. Group E, the experimental control, treated with iRNA 10 times. The dosage of iRNA was 0.16 mg/kg/day twice a week. After all treatments, five mice from each group were sacrificed on days 1,7,14 and 21. The splenic weight was evidently inhibited after radiation treatment, and iRNA could help the recovery of splenic weight. The thymus weights were significant loss after irradiation and a little increase in Group E. The recovery of bone marrow cells could be found in the groups which were treated with radiation. The phagocytosis in group E was highest among groups. The results indicated showed that the human spleen RNA seems can help the recovery of the immunity on immune organ and phagocytosis in the γ-irradiated mice. |
