中文摘要 | 台灣野生大豆G.tabacina族群分布於澎湖群島之原野的草原地,為多年生蔓性草 本植物,通常收取二年生以上之主根作為中藥,名稱叫做 " 一條根 "。 本試驗為瞭解種內 變異搜集澎湖群島八個地點的種子, 種植於台中中興大學,調查葉片、莢果及 DNA 多形性 。 小葉片及莢果的特性在種內族群個體間有顯著變異,但在八個地點間則無顯著差異。 以 30 條引子對所有樣本進行 RAPD 分析,在 30 條引子反應中,有 21 條引子產生 DNA 放大 產物,其中 11 條引子之產物具有多形性,共產生 79 條 DNA 的放大產物,內有 51 (64%) 保放大之產物具有多形性, 依條帶之出現計算各樣品間之相似度為 0.256 ~ 0.976 之間, 顯示在澎湖群島 G. tabacina 族群內有遺傳變異之存在。 |
英文摘要 | G. tabacina (Labill.) Benth. one of the wild soybean in Taiwan is distributed in the grassland of Penghu islets. It is a twining or porstrate perennial herb. Its main roots could be collected as herbs and was called "Yi-tyau-gen". In order to realize the intraspecies variations, the seeds from eight different sampling sits were collected in Penghu islets and planted in National Chung Hsing University, Taichung. Leaves, pods, and DNA polymorphism was investigated. The characteristics of leaflets and pods from eight different sampling sits showed no significant difference. However, intraspecies variations were significant. A set of 30 random sequence 10-mer primers was employed for all samples in RAPD analysis, of which 21 produced bands and 11 of these showed evidence of polymorphisms. Of 11 primers, produced 51 (64%) bands showed polymorphisms. Based on the appearances of markers, the genetic similarity coefficients were calculated and genetic variations were shown in the population of Penghus islets (0.236 ~ 0.976). |
