中文摘要 | 本研究以逢機增幅多型性DNA(RAPD)方法,探討蝴蝶蘭原生種分類及演化親緣關 係,結果顯示用16種蝴蝶蘭原生種為材料,以20種引子所產生之381條DNA條帶分析時, 其相似性係數及親緣順序呈現穩定,以此做為分類依據,可得到比形態分類更多更正確之資 訊,如形態分類上屬於遠緣之Phal. amabilis 與Phal. equestris,分析結果顯示其為相似一群, 另外,Polychilos節之Phal. mannii與Zebrinae節在形態分類上親緣關係亦相當遠,但 RAPD 分析顯示其與Zebrinae節之 Phal. lueddemanniana為接近一群,且與 Arends於1970年在細 胞遺傳學分析結果相符合。而由RAPD得到之樹狀圖配合Arends、Sagawa、Shino與Kamemoto 等人所進行核型分析資料進行比對,可知蝴蝶蘭染色體有由大向小演變趨勢,且親緣發展是 多源性。 |
英文摘要 | A random Amplified Polymorphic DNA (RAPD) analysis was used to understand the phylogenetic relationship of taxonomy and evolution among 16 wild species of Phalaenopsis. Results showed that stable similarity coefficient and relative order were obtained when 20 primers (381 DNA bands) had been analyzed. More reasonable accuracy of taxonomic information is obtained from RAPD data than morphology. Both Phal. amabilis and Phal. equestris that classified into two far-related asections by morphology are recognized as the most closely related species based on RAPD markers presented here. Similarly, Phal. mannii and Phal. lueddemanniana, which are different from morphological evidence, are also considered to be a closely related species by RAPD analysis that conforms to cytogenetic analysis as described by Arends in 1970. In addition, the result derived from the comparison between dendrogram generated by RAPD and karyotype obsered by Arends, Sagawa, Shino and kamemoto, implied that the evolutionary tendency of Phalaenopsis chromosome size is from the larger to the smaller, and its origin seems to be polyphyletic. |
