中文摘要 | 台灣雲杉成熟胚可成功的誘導出胚原性癒合組織(embryogenic callus),在供試的3 種培養基中,以LP培養基中添加10μM2,4-D及5μM BAP可達到最高誘導比率(6.8%), 濃度減半之培養基則未能提高誘導率。使用相同之植物生長調節劑組合,可維持胚原性癒合 組織不斷的增殖並繼代培養。以30μM ABA處理後,可使體胚進一步分化而成熟。在30 μM□ABA之處理中,添加不同碳水化合物可顯著影響體胚成熟的過程,此種促進體胚成 熟的效應以6%蔗糖優於6%麥芽糖或3%蔗糖,而又優於添加3%之麥芽糖。如果蔗糖之添 加以濃度漸進遞增或遞減方式行之,則以首先(前4週)加入6%蔗糖後,接著(後4週)保持相 同濃度的處理有較佳之效果。蔗糖濃度在後4週減為3%時,成熟體胚提早呈現類似萌芽之 現象。蔗糖濃度從3%逐次增加至4.5%b再提高至6%之處理,則使體胚出現不同步成熟之 現象。聚乙二醇(polyethylenc glycol,PEG)在3%及6%的濃度時,均有促進體胚成熟之效果。 成熟的體胚在置入1/2GD-1培養基2週之後,才開始有完全萌芽之個體出現,但是總萌芽 率僅達8.9%(n=90)。發芽之體胚苗已移植在溫室中完成健化,並移入蔭棚下存活生長。 |
英文摘要 | Somatic embryogenesis was induced from the mature zygotic embryos of Picea morrisonicola Hay.. Among three basal media tested, the highest percentage of embryogenesis (6.8%) was obtained on LP basal medium supplemented with 10μM 2,4-D and 5μM BAP. Strength of basal medium (half or full) did not affect percent embryogenic response. Embryogenic callus was maintained on LPG basal medium containing the combination of plant growth regulators mentioned above. Somatic embryos were transferred to LPG basal medium with 30Μm ABA and sucrose or maltose for maturation. The embryos performed the maturation superiority with the order: 6% sucrose > 6% maltose or 3% sucrose > 3% maltose. Addition of polyethylene glycol (PEG) at 3% or 6%(w/v) in the maturation medium improved the percent maturation of somatic embryos. The mature cotyledonary stage somatic embryos germinated on half-strength GD-1 medium without growth regulators after incubation for 2 weeks. The germination percentage was only achieved at 8.9% (n=90). The regenerated plants were acclimatized in the greenhouse and survived. |
