中文摘要 | 烏魚(Mugil cephalus)胃粗酵素液分解血紅蛋白(hemoglobin),在pH2.5∼4.5有較大的活性峰,其蛋白�t屬酸性之蛋白�t,最適作用溫度在45℃。以腸道粗酵素液分解酪蛋白(casein),在pH7.5及9.0有二個活性峰,顯示腸道之蛋白�t屬pH中性及偏鹼性蛋白�t,發現有胰蛋白�t(trypsin)和胰凝乳蛋白�t(chymotrypsin),其最適pH為7.5;而羧基胜�齘t(carboxypeptidase)A、B和胺基胜�齘t(aminopeptidase)之最適pH則在3.5∼4.0左右。且測得烏魚胃中之pH稍偏酸性,場道之pH則偏於中性,模擬烏魚消化道對蛋白質之消化,以烏魚消化道精酵素液於pH3.5先分解40分鐘,再將pH調至7.5分解200分鐘,其效果最佳,但此和腸道之羧基和胺基胜�齘t之最適pH在4.0不太符合,此點有待再深入探討。烏魚攝食後4小時,腸道中之胰蛋白�t和胰凝乳蛋白�t達最高,羧基胜�齘t則在攝食後至6小時均持續上升。 |
英文摘要 | The maximal hemoglobin hydrolysis activity of the crude enzyme extracted from the stromach of Mullet (Mugil Cephalus) were found at pH2.5∼4.5 and 45℃. The caseinolysis of the crude enzyme extracted from the gut of Mullet, maximal protelytic enzyne activities were found at pH7.5 and 9.0 respectively. The trypsin and chymotrypsin activities were maximal at pH7.5. Those of carboxypeptidare A, B and aminopeptidase were at pH3.5∼4.0. An in vitro system imitating the digestive process, using crude enzyme extracted from the digestive tract to hydrolyze casein, proteolytic activity was evidently increased by first reacting at pH3.5 hydralysis for 40 mins followed by reacting at pH7.5. |
