中文摘要 | 本試驗採取由台灣本島不同地區、不同時間自發病豬體分離得到的假性狂犬病毒株,包括已部TP1株(1984)、P3株(1986),中部CN株(1985)、CHU1株(1986)、CS株(1989),及南部TNL株(1976)、PT株(1989)等共七株,進行限制酵素切割之比較分析。這些病毒株先經三次病毒斑純化後再增殖,由其核蛋白衣體中萃取出病毒基因體DNA;再與兩株已發表的在美國分離得到的假性狂犬病毒分離株〔UCD和Shope株〕的基因體DNA分別以五種限制酵素〔包括KpnI、BamHI、PvuII、SmaI及HindII〕做切割。產物經0.7%瓊魯膠體電泳展開後,吾人比蠅其限制酵素切割片段的數目、大小,以及限制片段移走分佈的圖式。結果顯示,台灣分離株與美國分離株病毒基因體核酸的限制酵素切割圖式有著顯著差異,而台灣分離株本身各株間亦存在不小的差異。本報告之數據將有助於各台灣分離株病毒基因體物理圖譜之建立。 |
英文摘要 | We have compared the genomes of pseudorabies virus(PRV) strains from Taiwan and the United States by the restriction edonuclease analysis. Seven PRV strains isolated from different outbreaks in Taiwan, namely TP1, P3, CH, CHU1, CS, TNL, and PT strains, were examined and the two United States PRV strains (UCD and Shope strains) were chosen as references. All of them were cloned three times by plaque packing method with Madin-Darby bovine kidney cells before their genomic DNAs were extracted. The purified viral DNAs were separately digested with five restriction enzymes, including Kpnl, BamHI, PvuII, Smal, and Hindlll, and the resuting fragments were electrophoretically analyzed by 0.7% agarose gel. The results showed cleavage pattern variations in either the number or in the size of restriction fragments which clearly distinguished the seven Taiwan isolates from the two United States isolates, as well as from each other. The results of this study indicate that application of restriction endonuclease analysis is useful for differentiation of PRV field isolates. In addition, our data will help construct the restriction maps of the genomes of Taiwan PRV strains. |
