中文摘要 | 本試驗係利用馬鈴薯卡地那之試管苗,以及克尼伯之種薯,分別以化 學誘變劑甲磺酸乙酯(EMS)及疊氯化鈉(SA)以及γ-ray予以處理。誘變劑濃度, 試管苗,RMS,採用0.2-1.0共五種處理:SA以0.25一0.5mM其三種處理。在種 薯方面,EMS則採用0.4-1.2等三種處理,SA則採用0.25-0.75mM等三處理。γ-ray 採用5-15Kr共三處理。分別觀察其誘變效果。其結果顯示: (1)在低劑量EMS處理的腋芽中,其萌芽率及存活率均比對照為高。但濃度達0.6 時,漸漸降低;至1.0時,僅剩46.67。在SA處理,則所有萌芽率與存活率均較對照 組為低。至於種薯,化學誘變處理中,出土率與成活率均未受影響。γ-ray處理 中,其萌芽率與成活率雖較化學誘變處理為低,但仍達96。但劑量增到15Kr,則 成活率降至10.17。 (2)腋芽經化學誘變處理三十日後的節間長與節間數,各處理均無顯著差異。只 有0.5mMSA處理較其他處理稍短。在種薯處理中,十日株高以SA處理為最高, γ-ray處理為最低。 (3)在M1表現型上的變異,無論株高,根重,地上部重等性狀均只在高濃度EMS 及SA或高劑且γ-ray處理下,與對照有差異外,其他差異不大。但外型上有許多 變異植株出現,如植株矮化,地上結球等。 |
英文摘要 | The chemical mutagens, ethylmethanesulfonate (EMS) and sodium azide (SA), also thegamma radiation were used to treat the shoot of potatoes, in order to find their mutageniceffects. For the axillary bud treatments, the concentrations of EMS are 0.2-1.0, totally with5 treatments; and the concentration of SA are 025mM-0.75mM, totally with 3 treatments. Asfor tuber buds, the concentration of EMS are 0.4-12, totally with 3 treatments, the concentration of SA are 0.25mM-0.75mM totally with 3 treatments, and the doses of 7 -ray are 5Kr-15Kr totally with 3 treatments. The results are susmmarized as follows: (1) When the axillary buds were treated by the lower concentration of EMS, the emergences and the survival rates were higher than the control. However, when the concentrationwas increased to 0.6, they started to decrease. The survival rate of 1.0 EMS treatment wasonly 46.67. In the SA treatments, all the emergence and survival rates were lower than thecontrol. As for gamma-ray treatments, although the survival rates of low doses were relativelylower than the chemical mutagens, they still kept 96. However, when the dose was increasedto 15 Kr, the survival rate droped to 10.17. (2) When the axillary buds were treated with various concentration of chemical mutagens, the lengths and the numbers of internodes were not significantly changed, only the treatmentof 0.5 mM SA showed a little bit shorter. As for 30-days seedling heights, EMS treatmentswere higher than SA treatments. In the treatments of tuber buds, however, the SA treatmentgave a highest IO-days seedling heights. The T -ray treatment was the lowest. (3) In the phenotypic variations of MI plants, all the major traits like plant heights, weights of top parts were changed only in the higher concentration of chemical mutagens and higher dose of r-radiation treatments. Some mutants which include dwarf plants and the plants with upperpotatotubers were also found. |
