中文摘要 | 自恆春外海1公區及基隆港外1公里水深1公尺的海水,在23℃以marine agar 2216各分離細菌100株。恆春海水的好氣性總生菌數為36.2CFU/ml共分出11屬,27種菌,其中以Pseudomonas putida佔21%,Vibrio alginoliticus佔14%,P. stutzeri佔8%V. anguillarum佔7%等較多;在這些分離株中並無同時具有產生dchitinases及proteases d roh tss . ky bs wvom wn d ayo roh mgp bf 446CFU/ml,共分出12屬,23種菌,其中以Moraxella cuiculi佔20%最多,其次為Neisseria sicca 16%,在這些菌中有一株Alteromonas haloplanktis(標示為K-12)及一株Vibrio alginolyticus會產生chitinases及proteases,但只有A. haloplanktisindole不會產生indole。本A. Haloplanktis K-12之最適生長溫度為25℃,最適生長pH為7.6;在培養36 小時所得之粗酵素液(CES)中caseinase之活性最高;但在12小時以後得到的CES中chitinase的活性會降低。比較A. haloplanktis K-12與本實驗室自土壤分離Cellulomonas flavigena NTOU 1水解蝦殼水解液的收率,前者為43.3%,後者為58.8%。 |
英文摘要 | Each one hundred bacterial strains were isolated from seawater sample (1 m under water surface) collected form 1-km-off the Herng-Chuen coastal line or from 1 km remote from the Keelung Port. The aerobic plate count (APC) in the seawater of Heng-Chuen was 36.2 CFU/ml, including 11genera or 27 species of bacteria. The predominant species comprised, in descending order. Pseudomonas putida (21%). Vibrio alginolyticus (14%), P. stutzeri (8%), V. anguillarum (7%), and so forth. Among them, no one strain could produce both protease and chitinase. From the seawater (APC was 446.0 CFU/ml) of Keelung, 12 genera including 23 species of bacteria were isolated. The predominant species were composed of, in descending order, Moraxella cunicui (20%). Neisseria sicca (16%). N. mucosa (14%), and so forth. Among them, a Alteromonus haloplanktis (coded K-12) and a Vibrio alginolyticus could produce protease and chitinase, but only A. haloplanktis did not produce indole. The optimal growth temperature and pH of A. haloplanktis K-12 in crude enzyme solution (CES) prepared after cultivation for 36 h exhibited the strongest; whereas chitinolytic activity decreased in CES PREPARED AFTER 12 H CULTIVATION. The yield of shrimpshell hydrolysate liquefied by A. haloplanktis K-12 was lower (43.3%) than that (58.8%) by cellulomonas flavigena NTOU 1 which was isolated form soil by our laboratory. |
