中文摘要 | 細胞外間質的堆積及細胞的增生乃各種腎絲球腎炎最常見的現象。之前研究結果顯示在體外實驗中鹼基纖維細胞生長激素乃相當強的刺激素,為探討前覆性及溶解性細胞外間質蛋白成分對老鼠腎絲球間質細胞增生之影響及細胞外間質與鹼基纖維細胞生長激素間之交互作用, 吾人藉著改良MTT測定法來評估間質細胞增生,發現在富含血清的培養基上,前覆性細胞外間質無論是第一型或第三型膠原蛋白或纖維蛋白在高濃度下(每公分見方1.56 毫克)皆會抑制間質細胞的生長,於較低濃度下反而會刺激間質細胞生長。溶解性細胞外間質蛋白於高濃度的情況下,只有第一型及第三型膠原蛋白會抑制間質細胞增生,而溶解性織維蛋白無此現象。在無血清的培養基中,高濃度(每公分見方 469 毫克)前覆性第三型膠原蛋白可減低鹼基纖維細胞生長激素對間質細胞的增生作用,而纖維蛋白無此抑制作用。這些結果顯示細胞外問質蛋白不但對問質細胞有負迴饋現象且某些問質蛋白會影響鹼基纖維細胞生長激素對間質細胞的增生作用。 |
英文摘要 | Extracellular matrix expansion and mesangial cell proliferation are frequent findings in various glomerular diseases. Previous studies have demonstrated that basic fibroblast growth factor (b-FGF) is a potent mesangial cell mitogen in vitro. To further elucidate the role of the ex racellular matrix in rat's mesangial cell proliferation and its interaction with b-FGF, we investigated the effect of immobilized (precoated) and soluble (externally added) extracellular matrix (ECM) proteins on the proliferation of rat's mesangial cells and the effect of b-FGF on mesanigal cells precultured in a high concentration (4.69 ug/cm2) of immobilized ECM proteins. By means of a modified MTT assay, we discovered that precoated type I (C I) collagen, type III (C III)collagen and fibronectin (FN) exerted an inhibitory effect on mesangial cell growth at a high concentration (1.56 ug/cm2) but had a proliferative effect at a lower concentration (peak stimulatory concentration of C I: 0.00156 ug/cm2; C III 0.0156 ug/ cm'; FN: 0.0156 ug/cm2). At a concentration from 0.3 to 30 ug/ml,soluble type III collagen inhibited mesangial cell growth, but fibronectin did not. Soluble type I collagen also inhibited mesangial cell growth at the concentration of 30 ug/ml. In serum free medium, the proliferative effect of basic fibroblast growth factor was attenuated by a high concentration (4.69ug/cm2) of the precoated type III collagen, but not by fibronectin. These results suggest that ECM components may not only exert a negative feedback effect on mesangial cell growth, but also modulate the mesangial cell response to growth factors. |
