中文摘要 | 近年來的研究顯示血管內皮藉著包血管內皮舒張素在內之血管活性物質以媒介或管制血管運動,因此目前已被認為是一種內分泌器官。如粥狀動脈硬化,糖尿病與慢性高血壓等疾病狀態以往也被報告過會引起血管內皮功能失調。急性高血壓復被認為可增強serotonin之冠狀動脈收縮作用並造成血管內皮型態學上的損傷。但各家眾說紛耘而缺乏一致之認同。本實驗以離體血管環檢視靜注phenylephrine造成急性全身性高血壓是否會對基態及acetylcholine激發之內生性-氧化氮形成顯著影響。 LNMMA為一種內生性-氧化氮之生成抑制劑,它造成基態血管張力顯著增加,但此等反應未被急性全身性高血壓所影響。而acetylcholine獲張反應亦未被急性全身性高血壓所改變。高血壓前後LNMMA抑制acetylcholine舒張反應的幅度亦類似。 因此吾等認為急性全身性高血壓並不會造成周邊和系統血管內皮功能失調。內生性一氧化氮於不同部位血管作用之異質性未受影響。 |
英文摘要 | Vascular endothelium has been demonstrated to modulate vasomotor responses and therefore being recognized as an endocrine organ capable of releasing vasoactive substances. Varieties of disease conditions such as atherosclerosis, diabetes and chronic hypertension can induce vascular endothelial dysfunction, acute transient increase in blood pressure has claimed to potentiate sertionin induced coronary vascosnstriction with morphologic damage. The present study evaluated the possible effect of transient systemic arterial hypertension induced by parenteral phenylephrine on basal acetylcholine (ACh)-stimulated endogenous nitric oxide (NO) release in isolated canine coronary and femoral arteries. Baseline tension increase included by LNMMA, an endogenous NO synthesis inhibitor, was not significantly altered following transient hypertension. Ach-induced vasorelaxation following transient hypertension was not significantly changed. The extent of LNMMA inhibition on Ach-induced vasorelaxation with or witout acute hypertensive stress were comparable. In conclusion, transient arterial hypertension does not cause endothelial dysfunction in isolated canine coronary and femoral arteries. Heterogeneity is present in the endogenous NO modulation of vasomotor responses in different vasculatures. |
