中文摘要 | 本研究旨在評估裝於透明帶(zona pellucide)內(+ZP)之8-細胞期兔胚之單一 和二枚胚葉細胞( 1/8,2/8 胚)於體外培養,以及被置入體內迄 72 小時之分裂發育率和 20 天之著床率; 並觀察 1/8 胚經海藻酸鈣( Ca-alginate )膠囊包埋( micorencapsulation;ME )後,該膠囊包埋物於體內之宿命。 於體外培養迄 72 小時結果 顯示,1/8 胚或 2/8 胚( +ZP )之每胚葉細胞分製 3 次或以上(≧ 3 )之分裂製發育率 ,分別為 63.3% 或 55.2%;其中囊胚形成率分別為 36.4% 或 34.5%。二觀察值在不同胚葉 細胞數組別間之差異不顯著( P>0.05 )。若將裸露或僅膠囊包埋之 1/8 胚( -ZP )置入 體內,於置入後 24 小時後,均未能被回收。而不論經膠囊包埋與否,1/8 胚 (+ZP),於被 置入母體內 12 小時後, 均能順利進入子宮腔內惟膠囊包埋組( ME+ZP )之膠囊物已大部 分消退, 僅可見其殘餘物。 包埋和未包埋二組之細胞分製(≧ 3 )發育率,於置入後 24 小時,(P<0.05 ),48 小時則為 83.3% 和 71.4% (P<0.05); 迄置入後 48 和 72 小時之 囊胎形成率,分別為 33.3% 和 71.4%, 和 83.3% 和 66.7%。 於移置後 20 天,僅在 2/8 胚( +ZP )於顯微處理後,立即( 0 小組)經由輸管或子宮角腔壁被置入母兔生殖道內者 ,發現有著床之胎兒;其置入胚之著床率分別為 42% (1/24) 或 8.7%(2/32)。 迄懷孕終了 ,經由子宮角腔壁置入者產出一頭外正常之仔兔。資訊提示,已分離之胚葉細胞於被置入體 內孕育前,將之重行置入空透明帶處理仍屬必要;僅以海藻酸鈣膠囊包埋之處理,並不能長 久維持胚葉細胞於母兔生殖道內之整形態。於本試驗同時証實,裝於透明帶內之 8- 細胞期 兔胚之二枚胚葉細胞( 2/8 胚 P ),於體內確可被育成仔兔。 |
英文摘要 | The early stage of mammalian embryos have been micromanipalated to produce the identical multiples successfully. Manipulated rabbit embryos with damaged coverings reduced the survical rate in vivo, and denuded rabbit embryos failed to implant when transferred to synchronized recipients. The purpose of this study was to evaluate the development of the single- and two- blatomeres (1/8 and 2/8 embryos) isolated from rabbit 8-cell embryos in vivo and in vitro by encasing into an enacuated zona pellucida (+ZP) or/and encapsulating with outer layer of Ca-alginate gel. The development rate ( ≧ 3rd cleavage division) of the single- and two- blastomeres (1/8 and 2/8 embryos ) encased in an evacuated zona pellucida (+ZP) were 63.5% and 55.2% after cultivating in vitro for 72 h, and their blastula formation were 36.4% and 34.5% (P>0.05), respectively (in trail 1). In vivo, the denuded or the encapsulated single- blastomeres (1/8 embryo) with Ca- alginate gel (ME-ZP) could not be recovered in does genital tract at 24 h after transfer. Both groups of encased single- blastomeres (1/8 embryo) into ZP with or without Ca-alginate gel (ME) successfully arrived in uteri at 12 h after transfer, although its coating gel had almost degraded in the recipient genital tract. The development rates ( ≧ 3rd cleavage division) of the single- blastomeres (1/8 embryos) encased in ME+ ZP and ZP groups were 50 % and 50% (P>0.05) for 24 h, and 83.3% and 71.4% (P<0.05) for 48 h, respectively. Their blastulation formation were 33.3% and 71.4% for 48 h, and 83.3 % and 66.67% for 72 h, respectively. None of the recipients in single- blastomeres groups (+ZP) showed any signs of implantation which were examined on Day 20 after transfer. Three of six recipients were pregnant in groups receiving the two blastomeres (2/8 embryos) encased into an enacuated ZP. The proportion of implantation were 4.2% (1/24) and 8.7% (2/23) for two different transfer with two-blastomeres (2/8 embryos) encased into ZP, respectively. It was concluded that the zona pellucida is essential for the in vivo norml development of the single- and two- blastomeres isolated from 8-cell rabbit embryos; the single- and two- blastomeres encased into an evacuated zona pellucida would improve in the blastulation formation of blastomeres in vitro or in vivo, However, we didn't find the application of microencapsulating with Ca-alginate gel have any detrimental effect on the development of isolated rabbit blastomeres in does genital tract. |
