中文摘要 | 丙型血管張力素(angiotensin III; ANG III)曾被報導與ANG II具相等強度的神經胜肽,本實驗目的即是要探討為量注射ANG II及ANG III到腦幹內之獨立束核(Nucleus Tractus Solitarii; NTS)和最後區(Area Posterma; AP)對心臟血管的作用及可能作用機轉,及其在腎因性高血壓中可能扮演之角色。本研究是以urethane麻醉後之雄性Sprague-Dawley(正常、腎因性高血壓(兩腎、單夾模式)及假手術對照組)大白鼠為實驗動物。首先為證實ANG III在中樞神經系統具有調節血壓作用,我們從側腦室注射ANGII、ANG III,發現與以前專家學者所報導的ANG II、ANG III在側腦室,有劑量依賴性的升壓、心跳減慢的反應相符合;接著更進一步探討微量注射ANG III在NTS和AP對心臟血管的作用及可能作用機轉。我們將ANGII、III微量注射到NTS、AP內,發現低劑量的ANGII、III有劑量依賴性的降壓、心跳變慢的作用、而且最大反應的濃度是9.6 pmol;然而,高劑量的ANGII、III(9.6、480 pmol)在NTS內,卻有升壓和心跳加快的相反作用;另外值得一提的是低劑量的ANGII、III在NTS、AP內,對於腎因性高血壓鼠及假手術對照組之間,兩者產生不同的心臟血管作用,同時均抑制腎神經活性。對於ANGII、III(9.6 pmol)在NTS、AP的降壓和心跳減慢的作用,皆會被其對抗劑所部份或完全抑制,但ANGII、III是否作用在相同接受器上,則有待更深入研究。綜合以上實驗結果顯示,ANGII、III在NTS、AP部位有相同強度的降壓、心跳減慢的心臟血管作用,可能是抑制了交感神經活性所致。雖然經由本實驗我們發現ANG III在心臟血管反應有確定的基本特徵出現,但對於ANG III所扮演的生理角色仍不是很清楚,有待進一步加以探討。 |
英文摘要 | Angiotensin III (ANG III) had been reported to be an equipotent neuropeptide as compared to angiotensin II (ANG II). However, the cardiovascular effects of ANG III in the brainstem nuclei are still not clear. In this study we investigated the cardiovascular effects of microinjection of ANG II, III into the area postrema (AP) and the nucleus tractus solitarii (NTS). Male Sprague-Dawley rats (normal, renovascular hypertensive (2-kidney, 1-clip) and sham operated groups) were investigated in this study. When low doses of ANG II, III were microinjected into both NTS and AP, a dose-dependent depressor and bradycardic effect were observed, and the maximal depressor effect were found in 9.6 pmol. As the doses increased, pressor and tachycardic effect was found when microinjected into NTS. A significant difference of the cardiovascular effects of ANG III were noticed in between renovascular hypertensive rats (RVH) and sham operated rats when high doses of ANG III were microinjected into NTS, in the meanwhile, renal nerve activity were inhibited. The cardiovascular effects of both ANG II and III can be partially or completely abolished after pretreatment of their selective antagonists. Furthermore, interesting finding was noticed that the cardiovascular effects of ANG II and III in the AP were more potent than in the NTS. These results may suggest that the cardiovascular effects of ANG II and III in brainstem nuclei are complex, and they need further characterization. |
