中文摘要 | 本研究利用轉殖人類β-類澱粉蛋白(Aβ)基因之秀麗隱桿線蟲CL4176及CL2120作為模式生物,評估茶葉成分Epigallocatechin gallate (EGCG)對基因轉殖線蟲麻痺率與線蟲體內Aβ生成之影響。研究結果顯示EGCG可減少基因轉殖線蟲CL4176因Aβ引起的麻痺作用,證實EGCG可有效抑制線蟲體內Aβ之毒性。以西方墨點法分析線蟲Aβ單體與寡聚體的生成,評估EGCG對基因轉殖線蟲體內Aβ沉積的影響,EGCG可明顯降低CL4176線蟲因溫度誘導引起的Aβ寡聚體生成量。而對於CL2120線蟲Aβ生成的作用,EGCG亦可同時減少CL2120線蟲體內Aβ單體與寡聚體的生成。因此,綜合上述試驗結果,顯示EGCG具有預防或治療阿茲海默症之潛力。 |
英文摘要 | Anti-Alzheimer effect of tea component epigallocatechin gallate (EGCG) was evaluated using human Aβ gene transgenic CL4176 and CL2120 strains of Caenorhabditis elegans. Results showed that EGCG reduced Aβ-induced paralysis in transgenic CL4176 strain of C. elegans; it revealed EGCG inhibited the Aβ toxicity in transgenic C. elegans. Aβ deposits in transgenic CL4176 and CL2120 strains of C. elegans were analyzed by western blot analysis. EGCG could decrease the deposits of thermally induced Aβ monomers in transgenic CL4176 strain of C. elegans. As for the transgenic CL2120 strains of C. elegans, EGCG also decreased both the deposits of Aβ monomers and Aβ oligomers in transgenic CL2120 strain of C. elegans. Based on the above results, EGCG has the potential to prevent/treat Alzheimer's disease. |
