中文摘要 | 本研究擬探討樟芝萃取物在探討樟芝萃取物誘發人類肝癌細胞凋亡反應與其相關訊息路徑研究,本研究以肝臟 HepG2 細胞進行體外培養,以流式細胞儀分析癌細胞之細胞週期、細胞凋亡以及使用西方墨點法分析細胞凋亡相關訊息路徑變化情形與 下游蛋白質的表現,以進一步探討樟芝萃取物抑制肝癌細胞之機轉。在添加不同濃度之樟芝萃取物 (5、10、20μg/ml)下,研究結果顯示經 flow-cytometry 分析發現樟芝萃取物明顯誘發細胞凋亡反應,顯示樟芝萃取物確實具有促使肝癌細胞細胞凋亡之能力,然後經西方墨點法分析後發現,樟芝萃取物亦明顯誘發細胞凋亡蛋白 caspase3/7/9、 Cyt-C 與PARP(Poly (ADP-ribose) polymerase)等 (p<0.05),本研究結果證實樟芝萃取物可透過誘發細胞凋亡蛋白質之含量來達到誘發肝癌細胞凋亡反應的效果。 |
英文摘要 | The anti-cancer effects by Antrodia camphorate extracts against liver cancer were never been systemic evaluated. The purpose of this study is to verify the inhibitory effect on liver carcinogenicity by Antrodia camphorate extracts. In this study, we used Antrodia camphorate extracts to inhibit liver cancer cell proliferation and tumor growth, and investigate the underlying molecular mechanisms. HepG2 cell line(human hepatocellular carcinoma cell line)was used in this study. Cell cycles were analyzed by Flow cytometry. Signal transductions were analyzed by western blots. The results indicated the amounts of caspase3/7/9, Cyt-C and parp and the apoptosis effects were dose-related induced (p<0.05). The results from these investigations will prove to be helpful for understanding the anticancer mechanics by Antrodia camphorate extracts and provide novel insights in controlling liver cancer in our population. |
