英文摘要 | Charcoal's capability of adsorbing toxin has been reported in many applications. The advantage of charcoal in blood purification is its ultra-high capacity for toxin adsorption. In plasma perfusion where separated plasma or whole blood contacts with adsorptive materials (charcoal in this study), it is worth conducting bioassay to verify whether the materials are non-toxic to blood components. In our study, we investigated the biocompatibility of bamboo charcoal for the aphaeresis application. The bamboo charcoal was obtained by carbonizing Taiwan bamboo at 850℃ in the furnace. The surface morphology of carbonized particles remained in multi-cellular structure (honeycomb-like) of bamboo, and the pore size was characterized in two major groups: (1) less than 10μm and (2) 30-40μm. The specific surface area of the charcoal particles, measured by BET method was 836-971 m2/g. h the biological assays, the cytotoxicity test of L929 fibroblast cells showed that the charcoal particles were minor toxic, yet RAW 264.7 macrophage cells cultured with the charcoal particles expressed no inflammatory reaction, as compared to control group (culture medium). Nevertheless, the hemolytic percentage of the particles was around 1.4-2.6% indicating the minor damage of the red blood cells. The adsorption behaviors of bamboo charcoal in aqueous solutions varied in different polarity of the absorbent. The bamboo charcoal particles can rapidly absorb lipophilic amino acid (18-19 mg L-phenylalanine per gram of the particles), while it can only adsorbed 7.76 mg albumin per gram of the particles. Therefore, it was concluded that the bamboo charcoal particles can preferentially adsorbed lipophilic organic compounds. |
