中文摘要 | 本研究以小花蔓澤蘭(Mikania micrantha)之莖、葉及全株等作為試材,經冷浸法或熱迴流萃取法取得其乙醇萃取液,且將其以乙酸乙酯分層,探討各分離部對常見病原菌類(金黃色葡萄球菌、枯草桿菌、大腸桿菌及綠膿桿菌等)之抑菌性,並評估其於酸鹼與溫度變化之抑菌效果,另利用氣相層析質譜儀(GC-MS)探討其乙酸乙酯可溶部具抑菌可能之主成分。各乙醇萃取液抑菌性結果對枯草桿菌較其餘菌類者佳,且於濃度0.1 mg/mL 下即具有抑菌性,抑菌圈直徑介於15.03 - 16.15 mm,其中以熱迴流萃取法之全株乙醇萃取液的抑菌性最佳,其對枯草桿菌者之最小抑制濃度為0.78 mg/mL。於 pH值5.0至10.0之環境下對枯草桿菌之抑菌性具穩定性,另由耐熱性試驗結果可知,經常溫至溫度121℃變化下其仍具有抑菌效果,即表示具有良好之熱穩定性。又小花蔓澤蘭全株以熱迴流萃取法之乙醇萃取液經乙酸乙酯分層後,各分離部的抑菌性結果,以紙錠擴散法測定得知,其可溶部於濃度0.1 - 25.0 mg/mL 對枯草桿菌之抑菌圈介於15.49 - 21.08 mm,且最小抑制濃度經菌液稀釋法測定結果為0.39 mg/mL,顯示其乙酸乙酯可溶部者之抑菌性較佳,將其乙酸乙酯可溶部經GC-MS分析結果顯示,烯類之雪松烯 (18.57%)、β-蓽澄茄油烯(23.28%)及薑烯(23.70%)等含量最多。 |
英文摘要 | Whole plant, stems and leaves of Mikania micrantha HBK were extracted by the cold soaking processes and the heat reflux methods with ethanol, and the extracts were delaminated with ethyl acetate separation. Antibacterial activity of the extracts against Staphylococcus aureus, Bacillus subtilis, Escherichia coli and Pseudomonas aeruginosa were examined. The effects of temperature and pH value on their antibacterial activities were also determined. In ethyl acetate method, the main compounds of each separated extracts were identified using the gas chromatography mass spectrometry (GC-MS). Antibacterial activity of various Mikania micrantha extracts against Bacillus subtilis was better than that against other bacterial species at the concentration of 0.1 mg/mL with the diameter of growth inhibition zone between 15.03 -16.15 mm. The whole plant extracts from the backset refined methods with ethanol (HW) showed the highest antibacterial activity against Bacillus subtilis with the minimum inhibitory concentration (MIC) of 0.78 mg/mL. Analysis of pH value from 5.0 to 10.0 on the antibacterial activity showed that the pH change did not influence the antibacterial activity of the HW. In addition, the temperature change up to 121 °C did not effect on the antibacterial activity of the HW, suggesting good thermal stability of the HW. After delamination with ethyl acetate separation, antibacterial activity of each separated portion of the HW was effectively against Bacillus subtilis by the paper disk diffusion method with diameter of growth inhibition zone of 15.49 - 21.08 mm, and the MIC of 0.39 mg/mL determined by the broth dilution method. The results indicate that the ethyl acetate soluble portion showed the best antibacterial activity. GC-MS analysis showed the highest concentration of himachalene (18.57 %), zingiberene (23.7 %) and β-cubebene (23.28 %) of the alkenes compounds in the HW. |
