中文摘要 | 由台灣的魚隻分離出的三種鏈球菌經16s rRNA定序分析加以確認菌種分別為:Enterococcus faecalis、Streptococcus iniae、Lactococcus garvieae。將此三株鏈球菌以不同組合方式製備成福馬林殺滅全細胞疫苗,以高滲透壓浸泡免疫方式對吳郭魚(Oreochromis sp.)進行免疫,並評估此等疫苗的相對存活率(relative percent survival; RPS)。免疫攻毒後之相對存活率為72.2-88.7%,且單一免疫與補強免疫的保護力之間並無顯著差異,而血清抗體力價可在免疫後第二週檢測而得,而到第五至六週有明顯上升。吳郭魚在免疫一週後,其血清的細菌殺滅能力並沒有明顯的上升,但是巨噬細胞的細菌殺滅能力確有明顯上升。免疫後一週進行攻毒,其入侵的鏈球菌在第三至六天後便無法分離到鏈球菌。本實驗製備的福馬林殺滅全細胞疫苗可以誘發保護效力清除早期數週入侵魚體之鏈球菌,即便免疫後早期的血清抗體力價仍然低。 |
英文摘要 | Three species of streptococci (Enterococcus faecalis, Streptococcus iniae and Lactococcus garvieae ) isolated from aquacultured fish of Taiwan were identified by 16s rDNA sequencing. The formalin-inactivated whole cell vaccines of these streptococci were prepared. Tilapias (Oreochromis mossambicus ) were vaccinated by hyperosmotic immersion to evaluate the relative percent survival (RPS) of these vaccine. Besides, macrophages and sera of vaccinated fish were evaluated including bactericidal activity and antibody titer. In tilapia, the RPS was 71.2-88.7%, and the antibody titer significantly raised during 5(superscript th) and 6(superscript th) week after vaccinated. After 1(superscript st) week post vaccinated, the antibody titer did not increase but the bactericidal activity of macrophage increased. There is no significant difference of protection efficacy between single vaccination and booster vaccination. These formalin-inactivated vaccines of streptococci elicited a protective response to clear off the invasive streptococci in fishes during early weeks, even though the antibody titer were still low. |
