中文摘要 | 研究目的: 傳統中草藥長久以來被認為具有抗肝癌瘤的功效。過去五個月來本計畫 執行衛生署基因體中醫藥科技專案計畫探討中草藥對人類肝癌腫瘤之療效, 運用基因體技術分析其作用基因及探討其可能機轉,並進一步探討這些作用 的相關基因層次機轉與細胞功能性研究。證實已有兩種藥材會藉由影響細胞 內重要的轉錄NF-kB活性。初步結果顯示,複方藥材中,包括黃連解毒湯與香蘇散最能有效抑制肝癌細胞的增生,此外也進一步分析其單方成份,分別 顯示,黃連與紫蘇葉能有效抑制肝癌細胞的生長與移動侵襲。 方法: 這些有效中草藥對腫瘤細胞之作用包括:使細胞生長停滯、抑制由血清 誘發之細胞侵襲作用。未來仍應進一步顯示降低細胞株產生之血管新生作用 進一步由蛋白質層次確認主要的作用基因之蛋白質。 結果與討論: 中醫中藥治療肝癌是從整體出發,有攻癌,有扶正,症狀改善較顯著, 副反應小,療效較緩慢持久,但也存在著消除瘤體作用遲緩,針對性差等缺 點。這個計畫的執行,對釐清這些中草藥主要組成分作用機轉是對於未來用 在臨床上的應用,提供非常重要的依據。目前結果顯示,黃連解毒湯與香蘇散對於抗腫瘤可能有功效,應在檢測其組成單方成分之功效,然而對於鑑定其組成單方更多與其它抗癌方面之作用機轉與如何更有效應用仍有很大的空 間,相信在中醫藥委員會繼續支持下,將使得國家所投資發掘中藥材治療肝癌,讓研究計畫更趨完備。 |
英文摘要 | Purpose: For centuries, an abundant panel of Chinese herb medicines has been vastly applied in curing liver cancer. Effective as they are, their comprehensive action mechanisms have not been well understood that promised good opportunity for developing more novel and potential effective approach for inproving current liver cancer treatment. The purpose of this work is to investigate the biological activities as well as learning the relevant mechanistic knowledge and recognize metastatic inhibition potential in a panel of Chinese herbal medicine with an eye on those capable of suppressing angiogenesis of hepatocarcinoma cells. The proposed genomic research utilizing the extracts of the available Chinese medicine herb should provide in-depth evidence-based information on them. The work is expected to broaden current methods in treating hepatocacinoma that are prevalent in populations in Taiwan and in South East China. Methods: We have treated hepatocarcinoma cells with extracts from various herbs including single herbs and the combined formula, investigate their biological activities and identify the genes associated with angiogenesis and metastasis. The methods included growth study, invasion analysis and NF-kB luciferase determination. Results and Conclusion: Based on the finding up to now, we have identified Huang Lian Jie Du Tang (Coptis & Scute Combination) and Xiang Su San (Cyperus & Perilla Formula) as well their corresponding single herb components Huang Lian and Zi Su Ye are effective in suppressing growth and cell invasion in hepatocarcinoma cells. Therfore, it can be reasonably concluded that the two combined formula herb drug possess a highly potential for liver cancer therapy, their action and the mechanisms as well as possible application are worth of further studying. |
