中文摘要 | 很多種類的刺激會影響樹突細胞的功能,並且引導後續T細胞免疫反應的路徑,例如像是病原體的成分、疫苗佐劑、引起發炎的物質和植化物等。根據以往生藥學的研究結果,我們試驗坊間宣稱具有免疫調節功能的藥草萃取物,包括金線蓮、咸豐草、昭和草、山藥和紫錐菊等五種。藉由這些藥草萃取物對人類單核細胞演化之樹突細胞(MDDC)的刺激反應來瞭解這些藥草是否可能具有細胞調控的功能。我們以初代培養的細胞外觀型態、吞噬能力和細胞表面CD分子標記的表現量等方式來評估人類單核細胞演化之樹突細胞的功能活性。結果顯示,金線蓮和昭和草的萃取物對MDDC的分化有阻礙或抑制的作用,而對細胞成熟無明顯的影響;咸豐草和紫錐菊的萃取物則對MDDC的分化和成熟都有活化的效果;山藥的萃取物對MDDC的分化有幫助,但是對細胞的成熟無顯著的影響。研究結果可看出特定藥用植物萃取物含有免疫調節的成分,這些成分彼此之間可能對樹突細胞具有加乘的作用。根據這些結果,我們認為以樹突細胞作為免疫功能分析系統,將可以應用作為系統性地評估或是篩選具有關鍵性生物活性的藥用植化物或萃取物。 |
英文摘要 | A broad range of stimuli including pathogen-derived products, adjuvants, inflammatory mediators and phytocompounds can influence the function of dendritic cells (DCs) and then impact upon the subsequent T cell responses. According an approach of pharmacognosy, we tested the fractionated plant extracts from Anoectochilus formosanus, Bidens pilosa, Crassocephalum rabens, Dioscorea batatas and Echinacea purpurea, which had some legend and claims on immuno-modulatory effects. Therefore, we try to understand the possible effect of herbal extracts on modulating the cellular function of human monocyte-derived DCs (MDDCs). The functional activities and their modification of MDDCs were investigated by evaluating the morphology, endocytosis activity and expression levels of CD markers in primary cultures of MDDCs. Anoectochilus formosanus and Crassocephalum rabens which can block or inhibit the differentiation process without exhibiting a significant effect on DC maturation; Bidens pilosa and Echinacea purpurea, which can confer a weak activating effect on differentiation and maturation of DC; and Dioscorea batatas which may confer a weak activating effect on differentiation of DC, but have no significant effect on maturation of DCs. Our results suggest that mixtures of specific medicinal herb extracts may contain certain immune modulators, and they can work synergistically towards multiple targets of the DC immune cell system. Based on these results, we suggest that the current DC-based functional assay system may have application for systematic evaluation and screening of key immuno-regulatory bioactivities of candidate medicinal phytocompounds or plant extracts. |
