中文摘要 | 在高同半胱胺酸血症(hvperhomocysteinemia)的動物模式中,可見動脈血管鈣化(vascular calcification)現象,且其鈣化程度與血液中同半胱胺酸(homocysteine, Hcy)含量呈正相關性。本文旨在探討Hcy誘發血管平滑肌細胞(vascular smooth muscle cells)發生鈣化與造骨蛋白(osteopontin, OPN)表現之關係“分別將從大鼠主動脈取得的初代血管平滑肌細胞與不同濃度的Hcy (0, 10, 20, 50, 100, 200 pM)培養72小時,結果發現隨著Hcy濃度的增加,細胞增生的現象明顯增加。細胞外的鈣磷離子含量,隨著Hcy濃度的增加而逐漸減少。然而,細胞內的鈣及磷離子沉積情形、與鹼性磷酸酶(alkaline phosphatase, ALP)活性,呈劑量依賴型增加。顯示Hcy促進細胞外鈣磷離子進入糸細胞內,與ALP活性增加有關。Hcy濃度(10μM)低於高同半胱胺酸血症的水平時,亦可見細胞增生與鈣化的發生。此外,細胞鈣化發生伴隨出現OPN mRNA顯著表現,可能與阻止細胞鈣化有關。 |
英文摘要 | Vascular calcification has been demonstrated in experimental animal models of hyperhomocysteinemia, and the degree was associated with plasma levels of homocysteine (Hcy). The aim of the present study was to investigate the effects of Hcy on calcification of cultured rat artery vascular smooth muscle cells (VSMCs) and examine the alteration of osteopontin (OPN) expression. VSMCs were incubated with 0, 10, 20, 50, 100, and 200 μM Hcy for 72 h. Results showed that cell proliferation was promoted in the presence of increased Hcy levels. Furthermore, Hcy markedly decreased the extracellular contents of calcium and phosphorus. Dose-dependent increases in intracellular calcium and phosphorus concentrations, calcium deposition, and alkaline phosphatase (ALP) activity were also noted. Additionally, Hcy strongly enhanced OPN mRNA expression compared to the controls. This suggests that cell proliferation and calcification were still seen in the presence of Hcy levels (10 μM) lower than those normally associated with hyperhomocystemia. Hcy significantly increased calcium and phosphorus uptake rates in VSMCs, and it was associated with elevated ALP activity. The upregulated OPN mRNA may increase the binding to hydroxyapatite, thus inhibiting cell calcification by Hcy. |
