中文摘要 | 牛傳染性鼻氣管炎( Infectious bovine rhinotracheitis, IBR)及牛流行熱( Bovine ephemeral fever, BEF )病分別增殖在 MDBK 及 BHK-21 株化細胞,IBR 病毒力價 每毫升達 10 TCID �t,BEF 為 10 TCID �t, 培養後收集添加 0.002 M BEI 不活化劑予以 不活化完全後, 將此兩種病毒混合並添加礦物油佐劑作成 W/O/W 多相乳劑, 試製成 IBR 及BEF 不活化混合疫苗供牛隻防疫用。不活化劑選擇試驗方面,分別以0.2% Formalin,0.1% Formalin 及 0.002% BEI 進行比較,經免疫及測試天竺鼠及小白鼠,IBRV 中和抗體幾何平 均力價( GMT )分別是 21,32 與 45,BEF 則為 23,28 與 54,證實 BEI 不活化劑表現 較佳。 而添加佐劑比較試驗方面,有 10% 磷酸鋁膠及礦物油( W/O/W )兩種,結果 IBRV 之中和抗體分別是 37,52;BEFV 則為 49,60,經比較顯示礦物油成績較好。 在田間應用 試驗方面, 還擇兩場合計 35 頭牛隻作測試, 經疫苗接種後二週,可獲得良好免疫反應, IBR 中和抗體幾何平均值上升至 70 倍及 35 倍。BEF 則達到 176 倍及 129 倍。本試製疫 苗穩定性佳,在 4 ℃可保存一年以上,依然維持其免疫效力。 |
英文摘要 | Infectious bovine rhinotracheitis (IBR) virus and Bovine ephemerval fever (BEF) virus were propogated in MDBK and BHK21 cell lines, the highest virus titers of IBR virus was 10 TCID �t / ml and BEF 10 TCID �t /ml respectively. For the inactivative agent select test, the two kinds of harvested viral fluid were inativatived with 0.2% Formalin, 0.1% Formalin and Binary Ethylene Imide (BEI). After immunization of guinea pigs and mice, it was found that the neutralized antibody geometric titer (GMT) against IBRV was showed 21,32 and 45, respectively; against BEFV the GMT were 23,28 and 54. According to the data of inactivation the better responses of GMT shown in treatment of BEI. With regard to adjuvant efficacy compare test, two combined inactivated viruses were incoporated with either 10% aluminum phosphate or mineral oil (W/O/W). IBR antibody GMT showed 37,52 and BEF were 49,60. Comparatively, the immunity response of mineral oil was superior. The field trial for this combined vaccine was performed at two farms, that 35 heads of cow were treated. Two weeks later second vaccination at A and B farms, the neutralized antibody geometric titer (GMT) of the IBR were 70 and 35 respectively, as for BEF the GMT were 176 and 129. The stability and immunogenicity of the bivalent inactivated vaccine were confirmed in the maintained test at 4 ℃ for one year. |
