中文摘要 | 以假性狂犬病病毒(pseudorabies virus,PRV)屏東分離株(PT株)接種感染臺灣白鼻貍貓(Paguma larvata taivana)後,取其組織器官和血液,進行組織病理學、聚合連鎖反應和位雜合反應之檢測,並比較其敏感性和特異性。在組織病理學檢查,可見到大腦、延腦有嗜酸性核內包涵體、神經膠質細胞增生及神經元變性,肺臟出血及肝臟有局部壞死。以聚合?連鎖反應(PCR)、南方雜合反應及原位雜合反應(ISH)均可從感染的組織中檢測到病毒核酸。在敏感性方面,以溴化乙碇(ethidium bromide)染色可檢測到100pg病毒核酸,若配合南方雜合反應則可檢測到10pg病毒核酸。在檢測組織病毒核酸方面,PCR可在大腦、小腦、三叉神經節、延腦、扁桃腺、肝臟、肺臟和脾臟中檢測到病毒核酸,而ISH只在三叉神經節、大腦和肺臟中檢測到病毒核酸,故聚合?連鎖反應比原位雜合反應之敏感性和特異性較高。 |
英文摘要 | Giving an injection to Paguma Larvata taivana with pseudorabies virus (PRV)-PT strain, PRV in the tissues, organs and blood of the Paguma Larvata taivana was detectable. The molecular-based techniques such as polymerase chain reaction (PCR), Southern hybridization and in situ hybridization (ISH) were chosen to detect PRV DNA, and their specificities and sensitivities on the detection of PRV were compared. The findings in histopathologic examination were the intranclear inclusion bodies, gliosis and neuron degeneration in the cerebrum and the myelin cephalon, the hemorrhage in lung and the local necrosis in liver. The PCR and ISH could detect the PRV DNA from the infected tissues and organs, such as cerebrum, cerebellum, myelin cephalon, trigeminal ganglia, tonsil, liver, lung and spleen, indicating PRV can replicate in Paguma Larvata taivana. The PCR test was found to be more sensitive than ISH and could be detected. The sensitivity of the PCR as little as 100 pg of PRV DNA. The sensitivity increased 10 folds when PCR in combination with Southern bloting hybridization was performed. |
