中文摘要 | 我們篩選由中興大學化工作張傑明教授提供一系列松針萃取物,發現埮松松針萃取物(RPS與FP)對人類血癌細胞株HL-60、U937、K562與老血癌細胞株WEHI-3之生長具良好抑制作用,且抑制作用呈劑量依存性關係。初步的研究顯示赤松松針萃取物抑制癌細胞長生的作用可能經由細胞週期抑制及誘發細胞凋亡。動測試赤松針萃取物Balb/c小鼠之毒性,結果顯示口服給予赤松松針萃取物100mg/kg及1000mg/kg並無明顯的毒性反應。我們評估赤松松針萃取物在WEHI-3/Balb/c小鼠活體的抗癌的活性發現,經由WEHI-3癌注射之小鼠於28天後體重有明顯下降的現象,同時活動力降低,毛色光澤度差,有身體捲曲之現象。而無論是以WEHI-3癌細胞注射之小鼠同時口服給予赤松松針萃取物是給予癌細胞7天後再口服給予赤松松針萃取物的組別,雖然體重無明顯下降,其餘脾臟及肝臟腫大的情形,表面抗原增加的趨勢並沒有明顯的降低,因此推論赤松松針萃取物對WEHI-3/Balb/c小鼠體內血癌病變無明顯抑制作用。而由老鼠體重、活動力、肝臟及脾臟腫大情形,以及血清生化檢測值等毒性試驗結果顯示赤松松針萃取物對Balb/c小鼠無明顯的毒性,未來或許可從增加抵抗力等保健的方向作進一步的探討。 |
英文摘要 | We have investigated the effects of pine needless extract RPS (extracted from Pinus denisflora), on human leukemia cells HL-60, U937, K562 and WEHI-3. Adose-dependent decrease in cell proliferation was observed in RPS treated cells. The IC50 of RPS was 166.07 μg/ml for K562 cells respectively. Cell cycle analysis showed that RPS induced significant G0/G1 arrest and apoptosis in HL-60 cells. The RPS induced-G0/G1arrrest occurred before the onset of apoptosis. In animal study, RPS has less cytotoxitic effects in Balb/c mice on 100mg/kg and 1000mg/kg (P.O.). No effect of RPS on survival arte and body weight loss in leukemia mice with one month of experiment. The enlargement of spleen and lymph nodes and liver metastasis were not reduced in RPS treated groups. But flow cytometric analysis of leukocyte subtypes in PBMC showed that PBMC cell population was not change in RPS-treated mice than that in untreated mice. The results indicate that the RPS at concentrations of at 1000 μg/ml has no significant toxicity to Balb/c mice. These results suggest that PRS could provide as a “health-promoting” substance for human in the future. |
