中文摘要 | 本文研究臺灣原生蕙蘭屬植物金稜邊種子發芽形態,金稜邊蘭種子為紡錘形,長度569±50.3μm、寬度約160μm,種皮與胚之間為氣室,胚呈現極化,頂端細胞小,基端的細胞大,但未見胚乳、胚芽及胚根原體。種子基端具有開口,內外應可以通透空氣與水份。種子播種在培美基內一個月後膨大,胚頂端發育脹破種皮,形成黃白色原球體,照光後逐漸為綠色,種子發育成原球體的速度並不一致,播種後三個月可形成多量原體。綠色原長體為橢圓形,直徑介於1-2mm之間,表皮密生吸收毛。發育成熟的原球體頂端細胞較小,微凹陷,形成頂芽分生組織,基端胞較大,呈現尖形構造,未見後續發育。金稜邊種子離體培所發育形成的原球體,應為球形胚離開母體後發育成熟的胚。原球體後續發育有二種途徑,第一種可增生類原球體。第二種則從頂端分生組織形成莖葉,再長出根,發育成幼苗。 |
英文摘要 | Seeds of Cyumbidium floribundum Lindley are minute, dust-like, and fusiform in shape. They are 569±50.3μm long, 181.9±14.8μm wide, which have single-cell layer testa and an aperture in the posterior. Polar globular embryos are 290μm long, 160μm wide, and have small size cell in the anterior. There are no endorsperm, plumule and radicle primordium structures in globular embryo stage. Seeds of Cymbidium floribundum Lindley were sown in vitro and germinated to produce protocorms one month later. The absorbing hairs were initiated from the surface of elongated protocorms. The anterior of protocorm which was 1-2 mm in diameter produced the sheath leaf and apex primodium. It is a continuous development process from seeds to protocorms, and the protocorms could be considered as mature embryos. The protocorms developed further two distinct morphogenic routes, including: (1) proliferation of protocorm like-bodies as secondary somatic embryogenesis; (2) plantlet establishment with shoot and root. |
