中文摘要 | 如何有效自人或動物骨髓中分離MSCs在體外expand並調控誘導分化是相當值得探討的。另外,MSCs如何有效地在組織缺損無法修復或再生時植入達到修復或再生效果,是本世紀基礎與臨床再生醫學研究重要研發方向。因此,本研究旨在進行純化骨髓間葉幹細胞、增生和保存。研究結果發現,除了成功地利用比重梯度分離法和CD maker篩選法等進行人骨髓間葉幹細胞的純化和冰凍保存外,尚可在誘導因子處理6天後,看見具有脂肪小滴的脂肪細胞分化情形。並且隨天數增加,脂肪細胞數目、脂肪小滴大小均有明顯增加情形。因此,本研究結果顯示人骨髓CD34間葉幹細胞可被分離,純化和誘導分化。誘導其分化成脂肪 (或骨生成) 細胞等潛能。 |
英文摘要 | How to expand MSCs and direct them into different lineage cells of bone is important in regenerative medicine. In addition, transplantation of stem cells from bone marrow may play an important role in regeneration of bone defect. The purpose of the study was to isolate, purify, expand and induce differentiation of bone marrow stem cell. Six days after induction, lipid-droplet continuing adipogytes were noted. In addition, accumulation of lipid into a larger droplet and an increase in number of adipocytes was found with time. It is concluded that human CD 34 marrow stem cell could be isolated and differentiated after induction. |
