中文摘要 | 研究目的在探討傳統抗風濕中藥對組織單核球,滑膜纖維母細胞和內皮細胞分泌 附著分子及附著分子在這類細胞表面分佈的影響。包括組織單核球,滑膜纖維母細胞和內皮 細胞分別與不同濃度的傳統抗風濕中藥雷公藤、漢防己、皂刺、有骨消、黃金桂、柳枝廣、 大丁黃、西河柳、或昆明山海棠培養,並經第一介白質刺激後,以酵素免疫分析法測定其所 分泌的 E-selectin,ICAM-l 和 VCAM-l。 而細胞表面的附著分子則以流式胞儀測定。結果 顯示,高濃度雷公藤可抑制這類附著分子的分泌和分佈,其他中藥則無相似效應。但若將皂 刺、有骨消、黃金桂、柳枝廣、大丁黃、西河柳合併使用,則內皮細胞上的 ICAM-1 會略微 下降,但並無統計學意義。白血球和內皮細胞在發炎反應中皆扮演活躍角色,因之在發展新 一代抗發炎藥物的過程中,此二類細胞所帶有的附著分子即成為理所當然的目標。我們的結 果也顯示雷公藤在治療發炎性疾病上有其潛在價值。 |
英文摘要 | Objective. To observe the herbal medicine effects on both the secretion of adhesion molecules � from macrophages, dibroblasts, chondrocytes, and endothelial cells, and the expression of adhesion molecules over these cells. endothelial cells were incubated with different concentrations of Tripterygium Wilfordii Hook-f (TWH-f) or Tetradrine-in the presence or absence of interleukin-l (IL-1). The cell secreted E-selectin, intercellular adhesion molecule-l (ICAM-1) and vascular cellular adhesion molecule-l (VCAM-1) were determined by ELISA. The cell surface expression of adhesion molecules were detected by flow cytometer. Results. TWH-f at high concentration has a significant inhibitory effects on both the secretion and the adhesion molecules. However, Tetradrine did not demonstrate the same effects. Conclusions. Leukocytes and endothelium both play active roles in the pathogenesin of inflammation. The adhesion molecules of the endothelium and leukocytes may constitute excellent targets for the development of new anti-inflammation medicine. Our results indicate that TWH could be a potential therapeutic agents in the treatment of inflammtory disease. |
