中文摘要 | 人類流行病研究指出:快和慢乙醯轉移酵素與大腸和膀胱癌有關,也就是說大腸癌的起因中,此乙醯轉移酵素活性是非常重要的危險因子。本研究利用40位大腸癌病患的大腸腺癌組織來檢測此酵素的活性,結果發現可將此40個不同大腸腺癌組織,分成22個快乙醯轉移者,12個中間乙醯轉移者和 6個慢乙醯轉移者。生化特性的分析,此酵素最適宜的反應酸鹼pH值為7.5,最適宜的催化溫度為37℃此酵素易受 odoacetamide 的抑制作用。此酵素的分子量為34KDa。這是第一次研究有關中國人大腸癌組織中乙醯轉移酵素的特性的報告。 |
英文摘要 | Investigated for NAT activity toward p-aminobenzoic acid and arylamine carcinogen 2-aminotluorene. NAT activity toward these substrates indicated that all 40 of these colons could be segregated into 3 distinct phenotype groups. The patients could be subgrouped into rapid (22), intermediate (12). and slow (6) acetylators. The optimal pH value for the enzyme activity was 7.5 for both substrates tested. The optimal temperature for the enzyme activity was 37 ℃ for both substrates tested. The N-acetyltransferase activity was inhibited by iodoacetamide. Under 0.25 mM iodoacetamide, the NAT activity was reduced by 50% and at 1.0 mM iodoacetamide the enzyme activity was inhibited by more than 90%. Among a series of divalent cations and salts tested. Ca�� and Zn�� were the two most potent inhibitors. The molecular weight of NAT from human colon adenocarcinoma was 34 Kilodaltons (Kda). This report is the first demonstration of acetyl CoA: arylamine N-acetyltransferase activity in human colon adenocarcinoma tissues collected from a group of Chinese patients. |
