| 英文摘要 | A cationic derivative of cholesterol, 3β [N-(N',N'-dimethylaminoethane)-car-bamoyl] cholesterol (DC-Chol), is used as a component of cationic liposomes for efficient DNA transfection. The synthesis of DC-Chol has been reported. A modification to improve the yield of DC-Chol is described. Dry methylene chloride instead of dry chloroform was used to dissolve cholesteryl chloroformate and N,N-dimethylethylenediamine (DMEDA). Cholesteryl chloroformate solution was added dropwise to DMEDA solution at 0℃. After reaction, the solvent was evaporated and the residue was recrystallized in absolute ethanol at -20℃. The final product was identified by TLC (single spot), melting point (107.5 ~ 108.5 ℃),□H NMR spectrum and elemental analysis. The yield of DC-Chol was 73.2%, much higher than previously reported (21.8 %). The modified method offers a productive yield of DC-Chol for use in cationic liposomes. |
