英文摘要 | The creatinase from Pseudomonas putida RS65 was purified 9-fold increased in specific activity when compared with the cell-free extract through successive purification steps involving ammonium sulfate fractionation, DEAE-Sepharose CL-6B and Sephadex G-150 column chromatographies. The overall yield of the purified enzyme was 62%. The purified enzyme was confirmed to be homogeneous, and the molecular weight was estimated to be 46, 000 by SDS-polyacrylamide gel electrophoresis. The optimal pH and temperature for the creatinase activity were 8.0 and 40 ℃, respectively. About 75% of the original activity still remained after incubation at 45 ℃ for 1 h. The creatinase activity was completely inhibited by Ag ��,Cd ��, Cu ��,Hg �� and PCMB. |
