中文摘要 | 以真菌Aspergillus terreus醱酵生產其二次代謝產物lovastatin中,本研究探討其生產培養基之最適化。實驗上,先進行26-3部分因子實驗,得知乳糖、酵母萃取物、KH2PO4、KCl與PEG2000等五個組成份對產物的生產濃度有顯著影響;而改變脫脂黃豆粉濃度的效應雖不顯著,但該成份卻不可或缺。再由陡升路徑實驗與五因子五水準中心混成設計,結果顯示乳糖/KH2PO4、乳糖/KCl、酵母萃取物/KCl、KH2PO4/KCl等二階交互作用亦具顯著影響,此實驗設計所求得生產槽培養基的最適化濃度為(g/l):乳糖,109.290、酵母萃取物,10.920、脫脂黃豆粉,1.500、KH2PO4,1.520、KCl,1.114、PEG2000,0.452,於此組成下,二階迴歸模式預測產物濃度為1.16g/l,而6重複之平均實驗結果為1.14g/l,約為培養基最適化前的產量的2.67倍;此外,也驗證本生產培養基最適化之實驗設計法。 |
英文摘要 | We investigated the optimization of the production medium used in the fermentation of Aspergillus terreus for its secondary metabolite production. A26-3 fractional factorial design was initially applied, in which five ingredients, including lactose, yeast extract, KH2PO4, KCL, and PEG200, were found to significantly affect the titre of lovastatin in broth. Although the effect of defatted soybean meal was not significant, this ingredient was found inevitable in the medium for the production of lovastatin. We then conducted the experiments designed by the method of the steepest ascent and the central composite design. In the trials, there were significant interactions of lactose/KH2PO4, lactose/KCl, yeast extract/KCl, and KH2PO4/KCl on the system response. The work resulted in the optimal medium (g/l): lactose (109.290). yeast extract (10.920), defatted soybean meal (1.500), KH2PO4(1.520), KCl(1.114), and PEG 2000 (0.452). At the optimum, the regression model predicted a 1.16g/l of lovastatin meanwhile an average of experimental results with six replication was 1.14g/l. Compared with a 0.43g/l of titre of lovastatin before the procedure, we obtained a 2.67-fold increase in titre of lovastatin. It also verified the experimental design used in the medium optimization. |
