中文摘要 | 丁酮為國內工業界上常用之有機溶劑,而尿中丁酮濃度目前已被建議為勞工丁酮 暴露之生物暴露指標。本研究中,將含有丁酮之尿液檢體以頂空技術(head-space technique) 進行前處理,再以配備火燄游離偵測器之氣相層析儀進行定量分析。研究結果顯示最佳之前 處理條件為加熱溫度80℃,加熱時間為60分鐘,同時添加之碳酸鉀為2.50克。六條同日 與異日之檢量線相關係數均高於0.997,而各濃度之全變異係數(pooled coefficient of variantion)均小於3.50%,檢量線濃度範圍為0.08至51.20μg/mL。當尿液樣品分別貯存 於4及-20℃時,其穩定性能維持一個月以上。本方法之偵測極限與可量化偵測極限分別為 0.020和0.065μg/mL。添加回收率為97.49±2.78%。現場勞工真實尿液檢體測試結果亦顯 示本分析方法確實能適用於工作場所勞工丁酮之暴露評估。因此本研究已成功的建立了一種 能提供勞工丁酮暴露評估靈敏、簡單且可靠之生物偵測方法。 |
英文摘要 | 2-Butanone is a widely used organic solvent in industries. Urinary 2-butanone has been recommended as a good biomarker of exposure for 2-butanone. In this study, urine samples containing 2-butanone was treated by the head-space technique, then was analyzed by gas chromatography combined with a flame ionization detector. The optimal results were obtained when the heating temperature was 80 ℃ , the heating time was 60 minutes, and when 2.5 gm of potassium carbonate was added in the head-space sample pretreatment procedure. The correlation coefficients of 6 inter-day and 6 intra-day calibration curves of 2-butanone were greater than 0.997 and the pooled coefficients of variance were less than 3.5 % over the concentration range of 0.08 to 51.20 μg/mL. Urinary 2-butanone was stable for at least one months at 4 and -20 ℃ . The limit of detection and the limit of quantitation were 0.020 and 0.065 μ g/mL, respectively. The recovery of 2-butanone was 97.49 ± 2.78 %. Urine samples from exposed workers demonstrated that the proposed method is applicable to real samples in field study. It is therefore concluded that the proposed new method can provide a sensitive, simple, and reliable method for the biological monitoring of occupational exposure of 2-butanone. |
