中文摘要 | 本研究報告共採得了粉枝藻屬七種和蠕枝藻屬一種,及兩株粉枝藻屬未定種,經 抽取其genomic DNA後用Operon公司(OPP-01至OPP-10)十種引子作RAPD分析,並經由NT-SYS 計算其相似性及其樹形。相似性由未定種Ⅰ和南灣的細粉枝藻的0.11至未定種Ⅱ和紅柴的硬 粉枝藻0.95。分析結果顯示,殖絲粉枝藻較近似於東方粉枝藻、對生粉枝藻和細粉枝藻,而 和鼓苞粉枝藻、硬粉枝藻的差異較大。另外未定種Ⅰ和未定種Ⅱ在外形上也有別於其他粉枝 藻,結果顯示其遺傳形質和鼓苞粉枝藻自成一群。不同科的蠕枝藻其遺傳形質相近於本屬的 硬粉枝藻。粉枝藻從糯枝藻科分南提升為獨立的粉枝藻科,但經本研究RAPD的分析結果顯示 ,蠕枝藻和粉枝藻應是親緣關係極為相近的藻類。同種而不同區域的藻種中,小琉球、海口 、南灣三個地方的細粉褸藻相似性局達0.65-0.79,種內的變異比馬尾藻屬及石花菜屬等種 內的變異小。而殖絲粉枝藻雖然變異較大(0.l4-0.69),但和前兩者相比也是在其種間變異 範圍之內。 |
英文摘要 | Seven species of Liagora, one species of Helminthocladia and other two unidentified species were collected from southern Taiwan seashore. The genomic DNAs of all collected plants were extracted for random amplified polymorphic DNA (RAPD) analysis using 10 Operon IO-mer primers (OPP-01 to OPP-10). The DNA band patterns amplified from RAPD of 14 tested plants were matched together and similarity coefficients among these 14 individuals were calculated via NT-SYS. The dendrogram shows the relationship among these species. The similarity coefficients among these 14 individuals varied from 0.11 to 0.95, L. sp. I to L. segawai and L. sp. Ⅱ to L valida, respectively. Our RAPD results show that L. farinosa is similar to L. orientalis, L. decussata and L. segawai and dissimilar to L. setchellii and L. valida. Two unidentified individuals are similar to L. setchellii and separated from other Liagora species. The individual of Genus Helminthocladia is related to L. valida. This result suggests that Helminthocladia is related to Liagora. The similarity among L. segawai geographic individuals collected from three sites changed little from 0.65-0.79. L. farinosa varied from 0.14 to 0.69 in three collected samples. The variations of intraspecies and interspecies in Liagora are smaller than that of Sargassum and Gelidium. |
