中文摘要 | 小蘗鹼是黃柏的主成分;本研究選擇了小蘗鹼為模式藥物來設計用以治療皮 膚利什曼疾病之經皮輸藥系統。首先,將小蘗鹼加入幾丁質親水膠體製成軟膏,再 探討其軟膏的理化性質與藥物的釋出情形;結果指出,幾丁質親水膠體的黏稠度隨 著乳酸或EDTA的含量增加而增加,且EDTA的影響更勝於孔酸;而小蘗鹼的釋出 速率與軟膏的黏稠度則呈反比的關係。在體外皮膚的穿透試驗中,由於小蘗鹼低 的油水分配係數,使其僅有少量的藥物量能穿透老鼠皮;為了增加小蘗鹼的經皮吸 收,故使用界面活性劑為穿皮促進劑,在此類促進劑中,benzalkonium chloride對於 促進小蘗鹼穿透老鼠皮的效果最好,此外,Tween 80可增加小蘗鹼在老鼠皮中的負 載量。 |
英文摘要 | Berberine is the main ingredient of Coptis spp. This study selected berberine as a model drug to design the transdermal delivery system for the treatment of cutaneous leishmaniasis. Berberine was incorporated into chitosan hydrogel to prepare ointements. The physicochemical properties of the ointments and the release profile of berberine were investigated. The results indicated that the viscosity of chitosan hydrogel increased accompanied with the increasing amount of lactic acid or EDTA. The effect of EDTA on the viscosity was greater than that of lactic acid. The release rate of berberine was inversely proportional to ointment viscosity. In the in vitro skin perfusion studies, only trace amount of berberine had permeated through the rat skin corresponding to its low oil-water partition coefficient. The surfactants were used as penetration enhancers to increase the percutaneous absorption of berberine. Among the enhancers, benzalkonium chloride was found to be the most efficient for berberine to penetrate through the rat skin. Besides, Tween 80 could increase the loading amount of berberine in the skin. |
