英文摘要 | N-acetyltransferase (NAT) activity was determined in pineal glands collected from different aged pigeons (Columba livia) using 2-aminofluorene and p-aminobenzoic acid as substrates which were assayed by high pressure liquid chromatography. One-month-old or 6-month-old pigeons were sacrified during the light, during darkness or during darkness after a 1 min light pulse; in the latter case the pigeon's were killed 30 min after the 1 min light exposure. NAT activity in pineal glands from animals nocturnally exposed to 1 minute of light was inhibited in 1-month-old of animals. Nocturnal light exposure did not inhibit the enzyme activity in 6- month-old pigeons, even though these animals displayed clear light: dark differences in pineal NAT activity. Nocturnal light exposure also did not inhihbit night time levels of NAT activity in 1- month-old animals which had been bilaterally enucleated, suggesting that this effect is retinally mediated. Pre-treatment of 1-month-old and 6-month-old birds with a beta-noradrenoreceptor agonist, isoproterenol, prevented the nocturnal light-induced suppression of NAT activity. In experiments comparing the sensitivity of 6-month-old and 1-month-old birds to different illuminances or durations of nocturnal light exposure, it was found that the duration or intensity of light exposure needed to suppress nocturnal NAT activity was at least 4- to 5-fold greater in 1- month-old birds as compared to that seen in the 6-month-old animals. Kinetic constants for arylamine NAT activity in the pineal gland from animals with intermediate acetylator activity were determined. Km and Vmax values for 2-aminofluorene were 4-fold higher for birds exposed to light than for those not exposed. In summary, these results suggest that the retina-pineal gland pathway appears sensitive to light-induced changes in 1-month-old birds; however, this pathway does not appear to function in the 6-month-old birds in a like monner. |
