中文摘要 | 初期以自行設計之壓克力生物濾床式反應槽來馴養所需之微生物,利 用密閉之新鮮豬糞尿混合液與以化學試藥配製成之溶液作為臭氣來源。生物濾 床式反應槽內充填混有活性污泥與泥土之滅菌過蛇木,經過三個半月在室溫下 馴養,定時採取進流與出流之氣體樣本,分別經由單點式氣體分析儀與氣相層 析/質譜儀(GC/MS)分析結果顯示對於硫化氫、氨氣、 、糞臭素、酚及對-甲 酚皆可觀察到經生物濾床處理前後各臭氣成份濃度皆有減低之現象。然而,由 所採取之豬糞尿混合液中所揮發出之硫醇類濃度極低以致無法監測。經重覆實 驗所得之臭氣成份減低趨勢與第一次實驗結果頗相近。由反應槽內分離出六株 菌經初步分析其中五株為格蘭氏陽性桿菌,僅一株菌為格蘭氏陰性菌。在進行 反應槽實驗之同時,亦由泥土中篩選到數株假單胞菌屬(Pseudomonas spp)可在 有氧狀況下利用酚與對-甲酚為唯一之碳源與能源。對於硫化氫之去除則利用 Winogradsky column之方式由泥土中篩選出含硫之光合成菌,經使用混合菌株 進行定量分析,6天內可將培養液中約25 ppm之S 完全去除。而氨之去除也 另外由活性污泥與泥土中分離硝化細菌,可將氨氧化為硝酸鹽類。由反應槽中 分離出之6株細菌利用血清瓶進行對於分解 與糞臭素之定性與定量分析, 結果有2株菌可使用 為碳源;另外有3株菌可使用糞臭素為碳源。 |
英文摘要 | An acrylic cylinder was modified to be an enrichment bioreactor to isolate desired deodorous microorganisms. The mixture of freshly collected pig manure and urine as well as the solutions of phenol/p-cresol and indole/skatole were the only carbon sources for the microorganisms immobilized inside the bioreactor. Mixture of soil and sludge were then mixed with sterile snake-shaped wood branches. After a three-and -a-half month enrichment period, concentrations of those odorous components, hydrogen sulfide, ammonia, indole, skatole, phenol, and p-cresol were observed to be decreased after passing through the bioreactor. However, mercaptans were not detected because of trace amount. The detection experiment was repeated three times, and the data showed the same degradation pattern. There were six bacterial strains isolated from the biofilm covered wood. Five of them were Gram- positive and the other one was Gram-negative. On the other hand, several phenol and p-cresol degrading strains were isolated from the soil. Most of them were Pseudomonas spp. and phenol and p-cresol can be used as the sole carbon sources aerobically. Photosynthetic sulfur bacteria were isolated by the Winogradsky Column from the soil. Data showed that 25 ppm S were consumed completely by the photosynthetic sulfur bacteria in six days. Two of the six cultures isolated from the bioreactor were identified as indole-utilizing bacteria. Three of the six cultures were identified as skatole-utilizing bacteria. |
