中文摘要 | 本研究自臺灣大學校園內土壤篩選得NTU-FC-8菌株(經鑑定命名為Pseudomonas aeruginosa NTU-FC-8),以d-limonene為培養唯一碳源,進行發酵轉化實驗,發現P. aeruginosa NTU-FC-8可生長於含d-limonene 0.2~5.0%(v/v)之培養基中,且以d-limonene 1.5%(v/v)生長情況最為良好。無機氮源硝酸銨0.75%(w/v)可幫助菌體生長,0.6ppm的硫酸 錳及鉬酸鈉對菌體的生長有促進的作用。P. aeruginosa NTU-FC-8對d-limonene之主要轉 化產物經氣相層析/質譜儀鑑定為perillyl alcohol,該菌株於37℃,120rpm下培養60小 時,可使d-limonene生成perillyl alcohol之轉化率達到2.9%。於菌體生長至恆常期 (stationary phase),再額外加添d-limonene做為生物轉化基質,轉化率可提高至3.8%。 另外,將生長至對數生長期(exponential phase)之P. aeruginosa NTU-FC-8製成靜止細胞, 再加入d-limonene進行生物轉化,於37℃,120rpm下培養24小時,可使轉化率達5.5%。 |
英文摘要 | Pseudomonas aeruginosa NTU-FC-8 was isolated from the soil of the National Taiwan University campus by means of an enrichment culture using d-limonene as the sole carbon source. This strain could grow well under d-limonene concentration 0.2~0.5% (v/v) and generated a major oxygenated compound, which was identified as perillyl alcohol by GC/MSD, indicating it could convert d-limonene to perillyl alcohol. The optimal growth condition for the strain could be obtained by culturing in a medium containing 1.5% (v/v) d-limonene & 0.75% (w/v) NH□NO□ at 37℃and 120 rpm for 60 hrs. In addition, 0.6 ppm MnSO□ & Na□MoO□ could promote bacterial growth. The conversion rate was 2.9% under the optimal condition as described above. Addition of extra d-limonene during the stationary phase fermentation period could elevate the conversion rate to 3.8%. Furthermore, when P. aeruginosa NTU-FC-8 resting cells were prepared from the exponential phase and used for the bioconversion, the conversion rate could even be elevated to 5.5%. |
