中文摘要 | 甘麥大棗湯為中醫臨床上常用於治療臟燥之方劑,首載於<<金匱要略. 婦人雜病脈症并論>>,由炙甘草、浮小麥及大棗等三味藥所組成,主治「婦 人臟燥。症見精神恍惚,悲傷欲哭,不能自主,呵欠頻發或失眠盜汗」; 現今藥理學研究證實具鎮靜及抗痙攣作用。本研究擬採雙向主動迴避式反 應(Two-way active acoidance)、黑白室(Black and white chambers)及上 升性十字型迷宮(Elevated plus-maze)之焦慮模式,來探討甘麥大棗湯之 藥效,並以diazepam、buspirone為正對照組。 甘麥大棗湯於0.3∼1.0 g/kg劑量下,可減少大白鼠在雙向主動迴避反應 之非條件反應次數及誤動作次數,而對條件反應次數則不影響;另可增加 小白鼠於黑白室模式下在白室之滯留時間,並減少在黑室之滯留時間;最 後,於上升型十字形迷宮下,可增加小白鼠在開放臂之滯留時間百分率、 開放臂進入次數百分率及減少小白鼠在封閉臂之滯留時間百分率,但對封 閉臂之進入次數則不影響。另由運動活性及電痛閾值之測定,甘麥大棗湯 於實驗劑量下並不具鎮痛活性,但在1.0 g/kg劑量下具降低運動量之作用。 由上述結果顯示,甘麥大棗湯於0.3g/kg具抗焦慮之活性,且與鎮痛、鎮靜 之活性無關,其作用型態與buspirone相似;而在1.0g/kg劑量下,其作用型 態與diazepam相似。 在黑白室焦慮實驗模式,甘麥大棗湯可增強低劑量5-HTP及合成抑 制劑PCPA之作用,抑制高劑量5-HTP 之作用;亦可增強buspirone及 ritanserin之作用。而在甘麥大棗湯進一步之生化研究,在0.3g/kg時可增 加腦幹內5-HIAA之濃度,並減少5-HT之濃度。 綜合上述結果,甘麥大棗湯於小劑量(0.3 g/kg)下具抗焦慮作用, 其作用機轉與增加腦內5-HT之turnover rate有關。 由上述結果顯示,甘麥大棗湯於0.3g/kg具抗焦慮之活性 |
英文摘要 | Kan-Mai-Ta-Taso-Tang (abbreviate as Km) is a most common Chinese prescriptions used to cure insomnia and mania by traditional physicians. It firstly was described in "Chin-K'uei-Yao-Lueh" and composed of Glycyrrhiza Rhizoma, Triticum Seed and Zizyphus Fruit. In the modern pharmacological studies, Km has sedative and anticovulsive effect. Therefore, we attemp to investigate the anxiolytic effects of Km by means of two-way active avoidance, black and white chambers and elevated plus-maze, and use diazepam and buspirone as posititve controls. Km (0.3-1.0 g/kg) could decrease response counts of unconditioned stimulus and intertrial crossing in two-way active avoidance task, increase the time spent on the black chamber and decrease the time spent on the white chamber in the black/white box test. Finally, Km could increase the percentage of the time spent on the open arm and the entries of open arm, but decrease the percentage of the time spent on the close arm in the elevated plus-maze. Furthermore, Km at any dosage didn't have analgesic effect, but at 1.0 g/kg could decrease the horizontal activity (sedative effect). So, the anxiolytic effects of Km at 0.3 g/kg could be not related to analgesic and sedative effect, and it effects are similar to those of buspirone. Then, the effects of Km at 1.0 g/kg could be similar to those of diazepm. Km could enhance the effects of low dosage (5 mg/kg) 5-HTP, PCPA, *- OH-DPAT and ritanserin, and attenuate the effect of high dosage (50 mg/kg) 5- HTP in the black/white box test. Furthermore, in the biochemical measurement, Km at 0.3 g/kg could decrease monoamines levels and increase its metabolites levels in the brainstem. From these results, the anxiolytic effects of Km at 0.3 g/kg might be due to the increase of the turnover rate of 5-HT in brainstem. |
