英文摘要 | The roles of type I and II topoisomerase in the replication and gene expression of pseudorabies virus (PRV) were investigated by using topoisomerase inhibitors. Our results showed that both camptothecin (topoisomerase I inhibitor) and ellipticine and VP-16 (topoisomerase II inhibitors) could block the multiplication of PRV when added at time of infection. Camptothecin inhibited the incorporation of radioactive thymidine into viral DNA at late phase of infection; however, VP-16 and ellipticine did not cause significant reduction of thymidine incorporation. This finding suggested that type I topoisomerase activity is essential for the synthesis of virus DNA. In addition, our results also demonstrated that both type I and type II topoisomerases are required for the viral gene expression. Taken together, these data provided evidence for the requirement of topoisomerase activities in the replication and gene expression of PRV genome. |
