中文摘要 | 土壤分離之枯草桿菌可以在營養性培養液中產生蛋白酵素。蛋白酵素最適宜之活性條件為pH7.5和溫度55℃。凝膠電泳法評估蛋白酵素分解大豆蛋白質顯示分子量在31 kDa以下之胜無法分解。蛋白酵素之產生和蛋白質分解是成負相關。大豆蛋白質之分解較乳清蛋白質差,但是用大豆蛋白質做基質蛋白酵素產量較以乳清蛋白質做基質時高。在酵素凝膠電泳中有六個蛋白酵素活性帶。分子量分別為69.17,55.92,42.67,37.1,25.34和22.86 kDa。蛋白酵素抑制物EDTA和PMSF分別抑制蛋白酵素活性為24.93和69.09%。 |
英文摘要 | Bacillus subtillis isolated from soil could produce proteolytic enzyme in nutrient broth. Crude enzyme from culture supermatant showed that the optimum pH and temperature for activity were pH 7.5 and 55°C, respectively. Using SDS-PAGE to evaluate protein degradation indicated that the molecular weight of soybean protein below 31 kDa showed a resistance to degradation by protease. There was a negative correlation between the amount of degradation of proteins by enzyme and the amount of protease production. The protein degradation of soybean was lower than that of whey, while the amount of protease production in soybean used as substrate was higher than that of whey. There were six activity bands on the zymographic gel. The molecular weights were 69.17, 55.92, 42.67, 37.1, 25.34, and 22.86 kDa. The protease inhibitors, EDTA and PMSF, inhibited approximately 23.93 and 69.09%, respectively, of total protease activity of supernatant. |
