中文摘要 | 絕對厭氣性真菌大部分被發現於反芻動物的消化道之中。到1997年3月已被發表的有五屬十六個種。 根據國外的許多研究顯示,厭氣性真菌對於傳統細菌或真菌無法利用的結晶性纖維更具水解能力。但在國內對絕對厭氣性真菌進行的研究尚不多見。本研究中運用改良自Joblin的分離、培養方式、Marvin-Sikkema培養基及Hungate的rolling-tube等技術,以梅花鹿、山羌、長鬃山羊等三種臺灣本土特有種動物,及臺灣黃牛、駱馬及水失的糞物作為分離源進行絕對厭氣性真菌的分離培養,並在梅花鹿、水牛、黃牛及駱馬的糞物中分離出七株絕對厭氣性真菌菌株。依據其形態特徵,並參酌目前以發表的文獻中所載之型態指標,將D-21、X-26、X-34、Y-15、Y-24、L-87等具有多鞭毛的動孢子、絲狀假根菌絲體等型態特徵的六株分離株鑑定為 Neocallimastix屬,其中D-21、Y-15、Y-24、L-87為Neocallimastix frontalis,X-34為Neocallimastix hurleyensis而X-26則有待更一步確認。此外另一株從水牛所分離而得的X-55分離株,因具有單一鞭毛之動孢子及菌絲體世代所具有的球莖狀假根而為Caecomyces communis。 |
英文摘要 | Obligate anaerobic fungi are saprophyte in gastrointestinal tract of the herbivores and five genus which contain 16 species have been reported since 1975. The obligate anaerobic fungi are able to possess cellulases against crystalline cellulose which can't be degraded by bacteria and aerobic fungi well. In this study, we modified Joblin's isolation methods and medium which was developed by Marvin-Skiema. The Hungate's rell-tube technology was also used in our experiment. Finally, we isolated seven isolates of anaerobic fungi from the feces of llama, and Taiwan native animals water buffalo, Formosan Sika deer and Taiwan yellow bos. The seven isolates were cultured on modified basal medium with several kinds of carbon sources such as glucose, filter paper and Avicel. Based on their morphological characteristics, poly-flagellated zoospores, filamentous rhizoid systems the formations of monocentric colonies, the fungi, D-21,X-26,X-34,Y-15,Y-24 and L-87, were assigned to the genus Neocallimastix. According to their detail morphological characteristics, such as the ways of zoospores releasing, D-21, Y-15,Y-24,L-87 were classified to Neocallimastix frontalis and X-34 was assigned to Neocallimastix hurleyensis. The X-26 need futher obserbation to make sure its classification. The monocentric isolate X-55, isolated from the feces of water buffalo, produced uni-flagellated zoospores and bulbous rhizoid. According to these morphological characteristics, X-55 were classified to Caecomyces communis. |
