英文摘要 | The objectives of this study were using primary hepatocytes isolated from rat as an experimental model, to investigate the effects of various concentrations and incubation time intervals of bakuchiol on cell viability, lipid peroxidation, GSH concentration and its related enzyme activities. According to the results of LDH leakage, 0.125 mM bakuchiol treatment did not lead to a significant difference as compared with the control and caused adverse effects on hepatocytes. However, there were significant differences (P<0.05) in LDH leakage for 0.25 and 0.375 mM bakuchiol treatments after 8,24 and 48 hr incubation times, and adverse effects on hepatocytes. According to the results of lipid peroxidation, the TBARS value of each bakuchiol treatment was significantly lower (P<0.05) than that of the control over the whole incubation time. This result indicated that bakuchiol protected hepatocytes from lipid peroxidation. In terms of the intracellular GSH concentration of hepatocytes, 0.125 mM bakuchiol treatment significantly induced hepatocytes to produce more GSH; it was helpful for antioxidation and detoxication of hepatocytes. Considering the GSH-related enzyme activities compared with those of the control, 0.125 mM treatment significantly (P<0.05) elevated the activity of GSH Rd and maintained high concentrations of intracellular GSH; however, 0.375 mM treatment decreased (P<0.05) the activities of GST, GSH Px, and GSH Rd of hepatocytes significantly. Under this experimental model, 0.125 mM bakuchiol treatment was beneficial for the antioxidative physiological function of hepatocytes. |
