中文摘要 | 在大豆根瘤菌的研究上,快生型大豆根瘤菌全世界僅在臺灣及大陸兩地發現。而 對其結瘤因子的誘導產生、分離、及其基本性質都尚未有詳細的研究報告。本實驗嘗試著分 離本地快生型大豆根瘤菌不同菌株的結瘤因子並行初步的純化,另一方面也分離美國慢生型 大豆根瘤菌之結瘤因子,分別添加於大豆幼根,用顯微鏡觀察比較其對大豆根毛的影響,並 且比較不同種菌產生的結瘤因子對其感染初期菌體與宿主根毛吸附效應之影響。實驗結果顯 示,我們能在根瘤菌生長初對數期以類黃素加以誘導,分離出來的粗抽物可以在沒有根瘤菌 的狀況下使大豆根毛捲曲,證實有結瘤因子的存在。在分子篩的分離下,可以分出幾個可能 存在結瘤因子的部分。此外,在本實驗中,我們亦證實了在粗抽物中有成分可以影響菌體對 宿主根毛的的吸附作用。其意義在於結瘤因子可能不只是根瘤菌進入根毛的主要關鍵,還可 能是專一性吸附的重要因子,它可以增加菌體吸附的時間和數量,大大增加根瘤菌感染大豆 形成根瘤的機率。 |
英文摘要 | Fast growing rhizobia symbiosis with soybeans has only been found in Taiwan and mainland china, but there has been no detailed investigation of the inducement, productionm separation and purification of the nodulating factors (Nod factors) of fast growing rhizobia till now. In our experiment, we tried to separate the Nod factors of local fast growing rhizobium (Rhizobium fredii) and Nod factors of typical rhizobium Bradyrhizobium japonicum. We added the Nod factors to the soybean roots and checked their bio-effectiency by means of microscopy. Then, we also compared the adsorption efficiency of rhizobia if we added the Nod factors in the infectious process. We found that we could use Naringenin to induce some products when rhizobia grew to the rarly log phase and the products had curly root hairs.
