中文摘要 | 利用16條隨機合成引子行隨機複製多型性DNA(Random Amplified polymorphic DNA,RAPD ),以 9 個落花生品種(系)做遺傳分析,共產生 223 個隨機複製多型性遺傳 標誌( RAPD marker ), 其中有 183 個分子標誌在品種(系)間具多型性( Polymorphism ),有 83 個分子標誌為特定品種(系)的特有標誌( Unique marker )。 本研究所使用的引子中,以 B10 與 B16 最具分析潛力,不僅具有很高的隨機複製多型性標 誌,而且可偵測到的多型性標誌與特有標誌亦很高。 另外,在分析的 9 個落花生品種(系 )中,有 5 個為臺中場從立枝仔地方品種中所選育之品系,而且臺中育 13 號與臺中育 16 號二優良品系已參加區域試驗。 本研究不但可找到鑑別上述二品系的遺傳標誌, 在臺中育 13 號方面有 8 個特有標誌,臺中育 16 號有 21 個特有標誌,此即顯示選育品系與立枝仔 地方品種具有遺傳變異; 且由樹狀圖的結果可知,臺中育 16 號在 9 個落花生品種(系) 中,具最大的遺傳變異。 隨機複製多型性 DNA 分析,具有簡便、易於操作及效率高等特性 ,應用在落花生品系間之品種鑑定確實有效。 |
英文摘要 | Nine peanut lines/varieties were identified throught random amplified polymorphic DNA (RAPD) assay. Sixteen 10-mer random primers were used in polymerase chain reaction (PCR). In total, 223 RAPD molecular markers were detected, of them, 183 markers were polymorphic and 83 markers were unique. In this study, two primers B10 and B16 revealed that they were very powerful to detect the genetic variation among nine peanut lines/varieties. Furthermore, five lines were evaluated and selected from the local variety Lichi-tzae at Taichung DAIS. Among above five peanut lines, TC13 and TC16 have been evaluated in district yield trial. Eight unique markers from TC13 and twenty-one unique markers from TC16 were determined by RAPD assay. It also indicates that these selected lines have obvious difference on molecular basis compared with the original cultivar Lichi-tzae through two generations of selection and purification. From the phylogenetic tree based on RAPD bands among nine peanut lines/varieties, TC16 was found to have an apparent genetic diversity. Therefore, it shows that RAPD technology can be applied as a rapid, easy, and sensitive technique for peanut genetic analysis. |
