中文摘要 | 本實驗以遺傳工程技術應用在假性狂犬病毒活毒疫苗之開發。首先利用假性狂犬 病毒的基因體當作活毒疫苗之載體, 並將其毒性基因 TK 及 gI/gE 基因適當之裁剪刪除, 再將可表現豬瘟病毒之主要抗原決定基的 E2 基因嵌入上述假性狂犬病毒之多價疫苗。所篩 選到的重組突變株初步經墨點雜合反應偵測, 再以南方雜合反應及 PCR 確認 E2 基因之存 在。最後,利用 BALB/c 小白鼠進行毒力試驗,以證實所篩選到的重組突變株的毒力已大為 降抵,適合將來作為安全活毒疫苗的使用。 |
英文摘要 | Applying recombinant DNA technology, multiple deletion mutants of pseudorabies virus and recombinant pseudorabies virus carrying the E2 gene of classical swine fever virus were generated. Preliminary screening was performed by dot blotting and several prospective clones were further characterized by Southern blotting. The existence of E2 gene in recombinant viruses was also confirmed by PCR. Results of virulence test by using BALB/c mice showed that our recombinant viruses did not cause neurological signs or death in these experimental animals, suggesting that they can be used in future safety test using pigs. |
