中文摘要 | 重複兩次試驗,每次試驗均有5組,第1和2次試驗中每組分別有5至7隻雞, 總計60隻無特殊病原來亨雞於孵化後均飼養於過濾空氣之正壓雞舍中。兩次試驗之每組雞分 別以傳染性支氣管炎(IB)、傳染性喉頭氣管炎(ILT)、傳染性華氏囊炎(IBD)或家禽理奧病毒 感染症(AR)死毒油質疫苗或生理鹽水注射免疫各試驗雞隻,於免疫後1、2、4、6或8週採血 進行新城雞瘟(ND)、產蛋下降症(EDS)、Mycoplasma gallisepticum (MG)或M. synoviae (MS) 之血球凝集抑制試驗和自行製作的簡單免疫墨點(SDIA)和MG、ND、IB、ILT、AR和酵素吸附 免疫試驗(ELISA),並計算出各組之血清抗體力價幾何平均值,以為評估之依據。結果顯示 ND、EDS、MG或MS HI試驗的特異性很高,幾乎不見有其他抗體之交叉反應;在八種本實驗 室自行製作的SDIA試驗套組中,僅IBD顯現出高敏感性和特異性,ILT和AR的敏感性低特 異性高,ND、MG和MS非特異性高,IB之敏感性甚低,均需加以改進。在五種ELISA試驗中, 僅有IBD之敏感性和特異性均高,其他ELISA套組尚須進一步的評估。 |
英文摘要 | Comparison of the sensitivity and specificity of the self-developed simple dot immunoassay (SDIA), enzyme-linked immunosorbent assay (ELISA) and hemagglutination-inhibition test (HI) in the detection of the 4 antibodies of the 4 viral etiological agents of infectious bronchitis (IB), infectious laryngotrachitis (ILT), infectious bursal disease (IBD) or avian reovirus (ARV) existing in the serum of 60 special pathogen free leghorn chickens in 2 consecutive trials, 5 groups per trial, 5 and 7 birds per group in Trial 1 and 2 respectively was conducted. Birds in each group was immunized with one of the inactivated virus of IB, ILT, IBD or AR emulsified with a completed adjuvant or saline, then bled at 1, 2, 3, 4, 6 and 8 weeks post immunization for running the HI of Newcastle disease (ND), egg-drop syndrome (EDS), Mycoplasma gallisepticum (MG) or M. synoviae (MS); the SDIA of ND, EDS, MG, MS IB, ILT, IBD or AR and ELISA of ND, IB, IBD, ILT or ARV. The DN, EDS, MG or MS HI test was highly specific with almost no cross reaction with the heterologous antibodies. Only the IBD SDIA provides both high sensitivity and specificity and ILT and AR SDIA assay provides low sensivivity but high specificity in the detection of its homologous antibody. The ND, MG and MS SDIA gave rather high cross reactivity to the heterologous antibody and the IB SDIA assay gave too low sensitivity, which all need to be further improved. Also only the IBD ELISA was high sensitive in the detection of its homologous antibody and high specifically in the exclusion of the heterologous antibody. The sensitivity and specificity of the other 4 ELISA were rather poor. |
