中文摘要 | 為建立豆科作物組織培養之體系,以利品種改良及其他有關試驗之進行,先選定最適之培養基配方與生長激素組合,再測試品種及部位別之效應。試驗結果顯示,供試四個豆科作物(八個品種)中,根部癒合組織之誘起以紅豆最容易,菜豆其次,大豆再次,碗豆最困難。各品種花藥Callus之誘致率均很低,比較起來,紅豆最高,大豆其次,碗豆再次,菜豆最低。就紅豆之各部位言,根部最易誘起癒合組織,依序為子葉、胚莖、花藥。至於各種培養基之效應,根部以MS及1/2MS最好,Knudson及VW較差;花藥則以B₅之誘致率最好,MS次之,其他配方則不理想。試驗中,偶會出現分化之植株,但確切之再分化條件,仍待進一步之研究。 |
英文摘要 | Four kinds of leguminosae including soybean [Glycine max (L.) Merr.]. adzuki bean (PHaseolus angularis wight), pea (Pisum sativum L.) and bean (Phaseolus vulgaris L.) were used n this experiment. Callus induction from root-tip showed the highest percentage in adzuki bean, droped in the order of bean, soybean ad pea. Yet, callus formation from anther was in the order of adzuki bean, soybean, pea and bean. The facilitation of callus induction from explants of adzuki bean was in the order of root tip, cotyledon, hypocotyl and anther in this study. The 1/2 MS or MS medium supplemented with 1 mg/1 2, 4-D, 1 mg/1 NAA and 1 mg /1BA favored root tip callus formation of adzuki bean. However, the suitable medium for anther callus formation and growth was Gamborg's B medium supplemented with 2 mg/1 2, 4-D and 1 mg/1 kinetin. |
